Initial therapy and screening brief
Evidence cutoff: September 12, 2026. Worldwide human research, with U.S. approval and availability emphasized. This selective starting brief includes same-day additions to twenty-five categories; each category has fuller trial notes and sources. Future milestones below had not occurred at the cutoff.
Return to the cover page · Edit the watchlist · Dated research checks
1. Whole-body MRI
Current position: the research question—whether regular screening improves outcomes in asymptomatic people—remains unresolved. Professional radiology organizations cite insufficient evidence of improved survival and concern about unnecessary follow-up. Finding an otherwise hidden cancer in an individual does not by itself establish a net benefit for a screened population. ACR; CAR policy.
The studies to follow: Prenuvo's HERCULES targets up to 100,000 participants, assessing diagnostic performance and downstream outcomes. Its prospective observational design is not a randomized survival comparison. UK Biobank has imaged 100,000 people and is inviting up to 60,000 back for repeat imaging; this is a major research resource, rather than proof that routine commercial screening helps. HERCULES; investigator trial abstract; UK Biobank.
Expected update: no reliable date for a definitive answer. Late 2026–2027 is a low-confidence estimate for incremental observational news, not a promised outcome readout. The most important change would be controlled long-term evidence measuring benefits, false positives, procedures, and overdiagnosis.
2. Galleri and organ-specific cancer screening
Current position: the pivotal NHS-Galleri trial has already reported. In more than 142,000 adults undergoing three annual rounds, it missed the primary endpoint of reducing combined stage III/IV diagnoses in the prespecified cancer group. Secondary findings favored fewer stage IV cancers in later rounds. Mortality benefit remains unproven. This is a mixed result, not a successful primary-endpoint trial. Trial-team results.
PATHFINDER 2 adds prospective performance evidence: among 35,878 participants, the sponsor reported a positive predictive value of 60.3% and all-cancer sensitivity of 39.3% over 12 months. The test can find cancers, but a negative result is not a comprehensive cancer clearance. These are conference/sponsor results, with full publications and longer follow-up important next steps. PATHFINDER 2 results.
Expected update: an FDA advisory panel is officially scheduled for September 23, 2026. Its deliberations, briefing documents and vote are the clearest near-term event; an advisory vote is not FDA approval. Additional NHS follow-up is planned, but no firm release date was verified. FDA meeting.
Same-day scope expansion: newer colorectal blood/stool tests and precancer detection are now included. Shield and Cologuard Plus have FDA approvals but different diagnostic performance; detecting cancer and detecting removable precursors are distinct endpoints. Repeat-round outcomes and completed colonoscopy follow-up are priorities, with no reliable near-term result date verified. FDA Shield assessment, BLUE-C stool-DNA study.
3. GLP-1s and related metabolic therapies
Current position: oral obesity treatment is already a reality in the U.S., with oral Wegovy and Foundayo/orforglipron approved. Higher-dose injectable semaglutide is also approved. The category now spans weight management and specific obesity-related conditions; evidence and authorization must be assessed indication by indication. Oral Wegovy approval; FDA Foundayo letter.
Pipeline priorities: retatrutide has large Phase 3 weight-loss results but remains investigational. CagriSema produces substantial loss but failed noninferiority against tirzepatide in REDEFINE-4. Zenagamtide/amycretin, VK2735, MariTide, and eloralintide broaden the approaches to appetite, amylin signaling, dosing frequency, and maintenance. The topic page distinguishes full papers from topline announcements and avoids comparing percentages from unlike trials. Retatrutide latest pivotal update; CagriSema comparative result.
Expected update: September 28–October 2, 2026 is the EASD conference window, an opportunity for news rather than a promised result for every drug. Sponsor targets include a CagriSema FDA decision in late 2026, VK2735 maintenance data in Q3 2026, and retatrutide U.S. filing in Q1 2027. A filing is not an approval. EASD dates; Novo program update; Viking July update.
4. Muscle-building and muscle-preserving drugs
Current position: this is now supported by randomized human evidence, although the reviewed drugs remain investigational for obesity-related muscle preservation. Bimagrumab, trevogrumab and apitegromab have reduced lean-mass loss during GLP-1 treatment. For example, EMBRAZE found approximately 1.9 kg less lean-mass loss with apitegromab added to tirzepatide among participants with 24-week DXA, but no notable physical-function difference. Preserved DXA lean mass is not equivalent to proven strength or long-term health benefit. EMBRAZE paper; BELIEVE paper.
Safety can materially change the assessment: COURAGE's trevogrumab/garetosmab/semaglutide triplet had more discontinuations and two deaths; the sponsor reported no established causal association. The full data and independent interpretation matter. Formal enobosarm development is included, while gray-market use is excluded. COURAGE results.
Expected update: sponsor windows point to late 2026 for SRK-439 healthy-volunteer Phase 1 topline and Q1 2027 for enobosarm PLATEAU interim body-composition results; final PLATEAU topline is targeted Q4 2027. The SRK-439 result will not itself establish GLP-1 combination efficacy. Scholar Rock August update; Veru update.
5. Longevity and healthspan trials
Current position: there are real human trials, but the reviewed programs do not establish longer human lifespan. Metformin evidence is mixed: MET-PREVENT did not improve walking speed in frail older adults; a July 2026 preprint in older adults with glucose intolerance reported improvements in some frailty and methylation measures, but not all frailty measures. Neither provides a general longevity claim. MET-PREVENT; 2026 preprint.
Rapamycin and senolytics also have small human trials with missed primary endpoints and exploratory findings. Their existing use for other conditions does not establish an aging indication. The notebook follows functional and disease outcomes alongside safety, rather than equating biomarker movement with rejuvenation. PEARL rapamycin trial; senolytic bone trial.
TAME and expected update: AFAR's official page still describes fundraising for the planned six-year, 3,000-plus-participant program. A funded launch and enrollment were not verified, so there is no credible TAME readout date to put on the calendar. The next useful developments are funded launch/registration, actual recruitment, and peer review or replication of existing studies. Official TAME page.
6. Localized fat removal
Scope: drugs for regional subcutaneous fat and visceral-fat reduction, including new indications and GLP-1 combinations. Fat beneath the skin, visceral fat around the organs, and liver fat are tracked as different endpoints.
CBL-514: a 108-person randomized Phase 2b trial reported MRI-measured abdominal-fat reduction of 20.3% at four weeks and 17.3% at 12 weeks after final treatment, compared with little change on placebo. Those figures concern the treated abdominal depot. High attrition, unblinded clinician assessments, common injection-site reactions, and limited follow-up temper the result. It has not established a treatment for loose skin. Full Phase 2b paper.
Caliway reports that Phase 3 SUPREME-01 enrollment has begun. It also has clearance for a Phase 2 tirzepatide-combination trial; human evidence for preventing weight regain is still pending. August development update.
Expected CBL-514 update: September 30, 2026 is an announced EASD presentation on visceral-fat findings; the human analysis and preclinical contributions need appraisal when the abstract is available. Pivotal results are provisionally a 2027 watch item: the older registry estimates May 2027 primary completion, which can change and is not a publication date. Presentation announcement; SUPREME-01.
RZL-012: its sponsor reported positive China Phase 3 chin-fat results on September 2 and targets a China filing by end-2026, plus a future body-contouring study. U.S. timing remains unverified; this is sponsor topline evidence, not approval. Sponsor report.
AYP-101: another formal chin-fat program, with published Phase 2 evidence and a completed 252-person Phase 3 trial. Phase 3 results are not posted; no firm release date was verified. Phase 2 paper; Phase 3 registry.
Tesamorelin: included for VAT reduction and new indications. Its approved use remains HIV-associated lipodystrophy. There is human research outside HIV, including a selected abdominal-obesity trial and a completed non-HIV fatty-liver trial. The latter's small posted results concern liver fat and do not report numerical VAT outcomes. No broader-indication approval or dependable next readout date was verified. FDA label; abdominal-obesity trial; non-HIV liver trial.
Full localized fat removal notes and program-specific timing.
Same-day subtopic expansion: Cellulite is now included within localized fat removal and body contouring, with its appearance endpoint kept separate from fat-volume reduction. Avéli has FDA clearance supported by a nonrandomized one-year septal-release study; bruising and persistent adverse effects matter alongside photographs of improvement. Qwo is historical context because its sponsor stopped production and sales after concerns about bruising and prolonged discoloration. A small July 2026 study adds evidence for PLLA injections, but does not establish comparative superiority or a U.S. cellulite indication. The existing CBL-514 cellulite program remains on the list. Future checks will look for comparative trials, longer follow-up, regulatory changes and combined-procedure studies, separating effects of fat reduction, skin tightening and septal release. No reliable next cellulite results date was verified. The parent fat-removal baseline is preserved.
Added-subtopic timing: Cellulite: no firm next readout verified; watch comparative and longer-term evidence and CBL-514 follow-on studies.
7. Broad cardiovascular prevention, lipid drugs and screening
Current position: meaningful developments include oral PCSK9 treatment and gene editing. Enlicitide/Lipfendra received U.S. approval for LDL lowering in July 2026. VERVE-102, now under Lilly, has early human evidence of durable LDL reduction, reaching 62% at the highest tested dose in a 35-person Phase 1b report. Its cardiovascular-event benefit and lifetime safety remain unproven. FDA enlicitide approval; VERVE-102 report.
The major recent setback: on September 4, Novartis reported that pelacarsen's 8,323-person Lp(a)HORIZON trial missed its cardiovascular-outcomes primary endpoint despite lowering Lp(a). The full results are needed before deciding why or extrapolating to olpasiran and lepodisiran. Separately, olezarsen's expanded severe-hypertriglyceridemia indication includes pancreatitis-risk reduction, a different clinical outcome from prevention of heart attacks. HORIZON topline; FDA olezarsen update.
Expected update: full HORIZON data at an upcoming congress, date not announced; VERVE-102 Phase 2 enrollment targeted by end-2026. Longer-range registry/sponsor estimates put olpasiran's outcomes primary completion in March 2028, lepodisiran's in March 2029, and enlicitide's outcomes primary completion in November 2029. These are not release promises. OCEAN(a); ACCLAIM; CORALreef Outcomes.
Full cardiovascular notes and timing sources.
Same-day scope expansion: CAC, CCTA and AI plaque assessment are included, especially randomized evidence that screening-guided care prevents events. Symptomatic diagnostic evidence does not establish benefit from screening healthy people. TRANSFORM tests plaque-guided prevention in higher-risk asymptomatic participants; its registry estimates completion in March 2029, without a promised readout date. TRANSFORM registry.
8. Alzheimer's screening and therapies
Current position: blood-based testing is becoming more accessible for evaluating people with cognitive concerns. Roche's Elecsys pTau217 received FDA clearance in August 2026, following Lumipulse in 2025. Their intended use is diagnostic assessment in the appropriate symptomatic population, not indiscriminate testing of healthy people. Elecsys FDA decision; Lumipulse FDA explanation.
Leqembi and Kisunla slow decline in early symptomatic, amyloid-confirmed disease; they do not cure Alzheimer's or establish prevention in healthy adults. Leqembi gained a home-start subcutaneous option in July 2026. Brain swelling/bleeding risk and monitoring remain important. Pipeline priorities include trontinemab's brain-transport approach and the tau-lowering drug diranersen; the latter's CELIA trial missed its primary dose-response endpoint despite favorable cognitive analyses. FDA Leqembi update; CELIA disclosure.
Expected update: no firm immediate diagnostic or therapeutic readout date was verified. Important prevention trials have estimated primary completion in November 2027 for TRAILBLAZER-ALZ 3 and December 2028 for AHEAD 3-45; TRONTIER 2 estimates completion in June 2028. Results usually require additional analysis and publication time. TRAILBLAZER-ALZ 3; AHEAD; TRONTIER 2.
9. Psilocybin and related therapies
Current position: the category now has several late-stage drug-development programs. Psilocybin is a research focus; the watch also covers LSD, psilocin-related products, DMT/5-MeO-DMT, and MDMA, keeping their mechanisms, indications and evidence separate. FDA finalized psychedelic clinical-investigation guidance in July 2026; this is a research framework, not approval of the drugs. The key study questions include effective blinding, expectancy, durability, adverse events, and the contribution of psychological support. FDA guidance.
Psilocybin / COMP360: Compass reports two positive Phase 3 studies for treatment-resistant depression and follow-up through six months. Its rolling FDA application is underway, with final submission targeted for Q4 2026. The company's first-half-2027 launch target is conditional on FDA approval and required rescheduling; neither has occurred in the evidence reviewed. Full trial reports and regulator review remain important for interpreting magnitude, durability, and safety. August 5 sponsor update.
Other priority programs: DT120, formerly MM120, is an LSD formulation with sponsor-reported positive Phase 3 depression and anxiety results. HLP003, formerly CYB003, is a psilocin analogue approaching a pivotal readout. GH001 and BPL-003 are separate 5-MeO-DMT formulations; VLS-01 is a DMT formulation. The MDMA/PTSD program is tracked separately, with its reported resubmission still needing direct regulatory confirmation. DT120 results; HLP003 update; MDMA reporting limits.
Expected update: the nearest announced category-wide event is the FDA public hearing on September 14, 2026, 12:30–4:30 p.m. Eastern. It concerns future supervised therapeutic use and is not a product-approval vote or trial readout. Sponsor windows target DT120 Panorama results in September 2026, and HLP003 APPROACH and VLS-01 Elumina results in Q4 2026, alongside COMP360's filing completion. FDA hearing; DT120 timing; VLS-01 timing.
Full psychedelic-therapy notes and program timelines.
10. SSRIs and newer antidepressants
Current position: SSRIs now have their own watch, covering new drugs/indications, long-term efficacy, sexual and emotional side effects, withdrawal, and better treatment selection. Other antidepressant mechanisms are included as clearly labeled alternatives or adjuncts. Psilocybin remains a separate category.
A concrete SSRI development: the 2026 ADOPT PGx randomized trial did not improve its primary three-month symptom outcome with genotype-guided prescribing, although six-month remission favored it. This mixed result warrants follow-up rather than a claim that genetic tests reliably choose the right drug. Published trial.
Broader pipeline: follow newer monoamine drugs, serotonin-receptor drugs, orexin-based adjuncts and approved alternatives such as Auvelity and esketamine. Approval and evidence depend on the indication: zuranolone's U.S. approval is for postpartum depression. Keep failed or discontinued candidates in the history, including navacaprant and aticaprant. Detailed evidence and regulatory sources are in the topic notes.
Expected updates: Psych Congress September 15–19, 2026 has announced esketamine and seltorexant presentations; these need not be new pivotal readouts. The DISCARD antidepressant-tapering trial has conflicting completion estimates within 2027, with recruitment status requiring an update. Other program dates are labeled when no dependable milestone was verified. Conference announcement; DISCARD registry.
Full SSRI and newer-antidepressant notes.
11. Joint preservation and cartilage repair
Current position: Follow drugs intended to preserve cartilage and mobility, with pain, physical function and avoided joint replacement assessed separately from imaging. Lorecivivint has mixed pivotal evidence. The TG-C ACTiVION-II trial missed its co-primary endpoints; a second trial remains important. Lorecivivint trial, TG-C sponsor topline.
Expected updates: Sponsor windows: lorecivivint EMA filing later September 2026; TG-C ACTiVION-I topline October 2026. No firm FDA decision or sprifermin pivotal-readout dates verified.
Full topic notes, evidence and timing sources.
12. Sleep apnea and sleep/wake therapies
Current position: AD109 offers an oral airway-neuromuscular approach to apnea, with physiological benefit and substantial treatment discontinuation in its published trial. Oveporexton/Orzeyful is already approved for adult narcolepsy type 1; its U.S. launch still depends on separate availability steps. Home detection and broader sleep/wake treatments are also in scope. AD109 trial, Orzeyful approval and launch statement.
Expected updates: AD109 FDA target February 28, 2027. Orzeyful: sponsor expected DEA scheduling within 90 days of August 5, then availability; no completed scheduling or launch verified. Alixorexton: no firm next readout verified.
Full topic notes, evidence and timing sources.
13. Liver disease, fibrosis and noninvasive assessment
Current position: Resmetirom and semaglutide establish treatment options for selected noncirrhotic MASH, while preventing cirrhosis complications remains an outcomes question. Follow FGF21 drugs, combinations, and blood/imaging assessment. Efruxifermin missed its primary 36-week fibrosis endpoint in SYMMETRY despite a favorable later result. FDA resmetirom evidence, SYMMETRY paper.
Expected updates: Resmetirom/ervogastat interaction study targeted Q4 2026 and Phase 2 in 2027; MAESTRO-NASH-OUTCOMES results targeted 2027. ESSENCE outcomes completion expected 2029. No firm next EFX or test-qualification date verified.
Full topic notes, evidence and timing sources.
14. Cancer vaccines and immune interception
Current position: The watch separates treatment and recurrence prevention from prevention of a first cancer. Intismeran has positive melanoma Phase 3 sponsor topline results, with numerical data pending. BioNTech terminated a separate colorectal autogene-cevumeran trial after futility and a survival imbalance; its pancreatic combination trial continues. Lynch-syndrome prevention work remains early. Melanoma topline, Colorectal trial update.
Expected updates: Intismeran full phase 3 data at an announced but undated international meeting; no verified filing-decision or pancreatic/Lynch readout dates.
Full topic notes, evidence and timing sources.
15. Autoimmune disease modification and immune-reset therapies
Current position: CAR-T has encouraging remission signals in small, selected severe-disease cohorts. Rese-cel is progressing through a registrational myositis program, while approaches that omit preconditioning and generate CAR-T cells inside the body remain less established. Track organ function, relapse, infection and treatment burden. Academic case series, Current rese-cel program and forecasts.
Expected updates: Rese-cel myositis data targeted mid-2027 and possible BLA in second half of 2027 (August 13 sponsor forecasts). No dependable near-term preconditioning-free/in vivo readout date.
Full topic notes, evidence and timing sources.
16. Non-opioid acute and chronic pain therapies
Current position: New peripheral pain targets could expand treatment options for acute and chronic pain. Suzetrigine/Journavx is approved for acute pain; chronic neuropathic uses require their own evidence. Follow comparator-controlled efficacy, function, durability and tolerability, and retain unsuccessful trials. FDA acute-pain approval, Current Vertex pipeline.
Expected updates: Vertex sponsor window: complete both suzetrigine DPN phase 3 enrollments and VX-993 phase 2 DPN enrollment by end of 2026. Enrollment is not a results date; no firm next comparative Tonmya/Journavx readout date verified.
Full topic notes, evidence and timing sources.
17. Kidney preservation and earlier CKD detection
Current position: Established SGLT2 outcome evidence provides a benchmark for new kidney-preserving therapies and combinations. FIND-CKD extends finerenone evidence into non-diabetic disease, with slower filtration decline but uncertainty around the kidney-only event composite. Track pending indication expansions, early detection and whether acting on prognostic tests improves outcomes. FIND-CKD paper, FDA prognostic-test assessment.
Expected updates: Finerenone type 1 diabetes/CKD application has FDA priority review; China non-diabetic-CKD application submitted. No firm decision dates verified. KDIGO non-diabetic-CKD treatment update underway without a firm publication date.
Full topic notes, evidence and timing sources.
18. Hearing and vision preservation/restoration
Current position: Separate inherited deafness, common acquired hearing loss and vision loss. Otarmeni is approved for a narrowly defined genetic hearing disorder; that does not establish treatment for ordinary aging/noise damage. PRIMA supplies human evidence for limited prosthetic vision in advanced macular degeneration, with surgical risks and an uncontrolled study design. FDA hearing-therapy decision, PRIMA study.
Expected updates: Rincell-1 registry estimates November 2026 start and July 2028 primary completion. Otarmeni FDA-required protocol deadline December 30, 2026, final confirmatory report August 30, 2030; no firm earlier public readout date. PRIMA next public clinical/FDA milestone undated.
Full topic notes, evidence and timing sources.
19. Hair loss and hair restoration
Current position: The aesthetic priority is pattern hair-loss prevention and visible regrowth. Follow PP405, clascoterone 5%, VDPHL01, KX-826 and GT20029; keep autoimmune alopecia separate. Most pipeline findings remain investigational or sponsor-reported. Compare absolute hair gains, visible benefit, durability and safety within each study. PP405 current plan, Clascoterone filing guidance, VDPHL01 readout windows.
Expected updates: VDPHL01 male confirmatory/12-month data: H2 2026; PP405 late-stage start: 2026, not verified started; clascoterone U.S. filing: early 2027 (sponsor targets).
Full topic notes, evidence and timing sources.
Same-day subtopic expansion: Hair graying and biological pigment restoration now sit within the hair category. Human studies establish that individual hairs sometimes regain pigment, but they do not prove broad, durable reversal of ordinary age-related graying. Repigmentation during treatment of autoimmune alopecia remains a separate context. BioQ describes BQ-002 as a sustained-release injectable candidate; no matched human trial, efficacy dataset or U.S. approval was verified. A May 2026 Reddit post predicts a Phase 1 start in 2027. The candidate exists on the sponsor website, but that forecast remains explicitly unconfirmed and will be followed as a rumor. The next meaningful milestones are credible trial registration, actual first dosing and controlled human results. Gray hair counts, visible coverage and durability will be distinguished from hair-growth endpoints, cosmetic staining and changes confined to individual strands.
Added-subtopic timing: Hair graying: no firm date verified. BQ-002 Phase 1 in 2027 is an explicitly unconfirmed Reddit claim, not official guidance.
20. Skin aging, laxity and regeneration
Skin aging and laxity now cover drugs, gene therapies, devices and procedures, with emphasis on visible and durable results after weight loss. KB304 has a small placebo-controlled Phase 1 signal for upper-chest wrinkles, but this does not establish long-term benefit or correction of substantial loose skin. Its latest sponsor timeline targets Phase 2 initiation in 2027, replacing the older 2026 plan. Established tretinoin evidence provides a comparison for newer topical claims. A 2026 randomized radiofrequency study found YOUMAGIC noninferior to Thermage; an active-comparator result does not establish superiority over sham. FDA is separately evaluating reports of serious complications with certain RF-microneedling uses. Track skin quality, objective laxity, downtime and unintended fat loss separately, and cross-link facial volume changes. Beyond the KB304 trial-start forecast, dependable results or regulatory-decision dates were not verified.
Expected updates: KB304 Phase 2 start targeted 2027 (August 2026 sponsor forecast); no reliable readout date. Device comparisons and FDA RF-microneedling review undated.
Full topic notes, evidence and timing sources.
21. Scar prevention, treatment and stretch marks
Scar research now includes formal human attempts to prevent surgical scars and remodel established ones. Geneva has registered a small verteporfin trial, but its old recruitment status and elapsed completion estimates need verification. Syntara is studying topical LOX inhibitors for hypertrophic scars and keloids; the current program supports tracking clinically visible improvement, symptoms and durability rather than assuming that tissue changes mean scar removal. A small randomized study of radiofrequency plus topical tretinoin for mature stretch marks found a patient-reported benefit without significant blinded-observer separation. Different scar types remain distinct subtracks. SNT-9465 results are targeted during 2026, while there is no dependable public date for verteporfin results or a definitive stretch-mark advance. Trials remain early, and their specific scar indications must be distinguished from existing approvals for a drug or device in another use.
Expected updates: SNT-9465 recruitment targeted Q3 2026 and topline during 2026 (sponsor forecasts); verteporfin and stretch-mark readouts have no dependable near-term dates.
Full topic notes, evidence and timing sources.
22. Facial volume restoration and contour
Facial volume restoration now includes fillers, collagen-stimulating injectables, fat grafting and new delivery approaches, especially after weight loss. The Sculptra-plus-Restylane study previously summarized as a 35-person trial now has a peer-reviewed paper: 41 people enrolled and 35 completed. The open-label study reported improved appearance and satisfaction through nine months after final Sculptra treatment, but cannot separate product effects or show superiority over other approaches. Regulatory status remains specific to each filler and anatomical indication; overseas expansions are not U.S. approvals. Fat-transfer innovations should be assessed by measured retained volume, revision needs and long-term appearance rather than selected photographs. Serious and delayed filler complications remain part of surveillance. There is no dependable dated pivotal readout or U.S. decision for this category in the baseline; the most useful next developments would be controlled studies and longer follow-up.
Expected updates: No reliable pivotal readout or U.S. decision date verified; watch comparative post-weight-loss trials, longer follow-up and indication-specific approvals.
Full topic notes, evidence and timing sources.
23. Pigmentation and uneven skin tone
This category covers unwanted dark pigmentation, with vitiligo kept separate because its aim is to restore lost pigment. A small malassezin study versus hydroquinone supports further investigation but does not establish equivalence. A larger controlled thiamidol trial improved melasma-severity scores while patient assessments and sunspot outcomes did not significantly separate, illustrating why a positive score should not become a blanket claim about dark spots. For vitiligo, topical ruxolitinib is already FDA-approved in a defined population, while oral povorcitinib has positive Phase 3 sponsor topline. Further povorcitinib data were forecast for the second half of 2026 and regulatory applications for the first half of 2027; these are development forecasts. The watch will prioritize lasting visible benefit, tolerability, relapse and results by skin type, while seeking new human programs for PIH and solar lentigines.
Expected updates: Povorcitinib additional vitiligo data H2 2026 and filings H1 2027 (April sponsor guidance); no firm next malassezin/thiamidol milestone.
Full topic notes, evidence and timing sources.
24. Acne and rosacea
The most distinctive early program is Sanofi’s SP0268 acne mRNA vaccine: its current sponsor page lists a recruiting Phase 1/2 study with 800 planned adults. No efficacy readout or reliable release date was verified. Emrosi provides a newer approved oral option for inflammatory rosacea, supported by randomized comparisons, but durable remission and broader redness benefit remain separate questions. DMT310/XYNGARI illustrates why sponsor updates matter: an encouraging acne trial was followed by a pivot toward cosmetic/OTC products, while its rosacea program had already been stopped after a negative trial. The category also covers formal device and procedure evidence, relapse, scarring prevention, skin-tone representation and treatment burden. Next milestones are a verified vaccine timetable and early data, meaningful comparative follow-up, and genuinely new human programs. Cosmetic launches will not be treated as drug approvals.
Expected updates: No dependable near-term readout date verified; reconcile SP0268 registry timing and watch initial efficacy, durability and new trial announcements.
Full topic notes, evidence and timing sources.
25. Dental alignment, regeneration and repair
Dental alignment is included explicitly: aligners and braces, direct-printed materials, clinician-supervised remote monitoring, treatment speed, root/gum safety, and retention after treatment. A recent small randomized study found that three-day trays with reduced programmed movement produced more refinements and visits without shortening total treatment compared with seven-day trays. An older remote-monitoring trial also illustrates why fewer appointments and faster treatment must be assessed separately. Recent alignment trial.
Regeneration covers early enamel lesions, periodontal support and missing teeth separately. P11-4 plus fluoride has small human orthodontic-prevention data; Toregem's TRG035 has reached Japanese Phase IIa trial-notification review for congenitally missing teeth. That milestone does not establish human tooth regrowth or an adult replacement for ordinary lost teeth. Enamel trial, TRG035 milestone.
Expected updates: TRG035 Phase IIa enrollment/dosing after August 2026 trial review; no firm dental readout date verified. Several orthodontic registry statuses need refreshing.
Full topic notes, evidence and timing sources.
How this baseline will be used
The Tuesday/Friday checks compare new developments against this snapshot and update the topic pages and cover page. Announced dates, sponsor targets, registry estimates, and analyst estimates remain visibly distinct. A missed or shifted milestone is itself worth investigating; a scheduled study-completion date is never treated as a result that has occurred.
The linked topic pages document coverage limits, including incomplete live registry rendering for the MRI/CBL-514 checks and reliance on labeled sponsor/conference reports where full results are not yet appraised. This brief is research tracking, not a recommendation to start a treatment or screening program.
