Home · Latest · Calendar

Worldwide human research with U.S. availability called out separately. All 28 active topics received focused discovery and source checks. Searches covered the seven-day overlap from retained successful-check dates, generally September 5–18 (September 6 for the newer September 13 baselines), plus named historical gaps. None of the 28 topics received an exhaustive complete-topic re-review: successful-check timestamps are unchanged. Unresolved gaps remain queued for the next run.

Findings and evidence corrections

Neuro-DORA studies sleep treatment and neurodegeneration biomarkers

Source date: 2026-09-18. Evidence: recovered baseline; current registry update.

New to this notebook, Neuro-DORA is an existing small trial of lemborexant and blood biomarkers, not a newly launched dementia-prevention trial. NCT06823752, Woolcock Institute Australia; 20 planned adults with insomnia, randomized masked crossover, two weeks lemborexant versus placebo separated by washout. Primary pTau181 ratio; pTau217, amyloid, NfL and GFAP secondary. Actual start May 13, 2025; September 18 record recruiting, no results.

Limitations: Brief biomarker experiment, not dementia incidence, cognitive function or durable insomnia benefit; update alone does not establish a substantive protocol change.

Safety: No results posted; do not infer neuroprotection from study rationale.

U.S. status: Lemborexant/Dayvigo is an insomnia medicine; this study does not add an Alzheimer-prevention indication.

Next milestone: Registry estimate: Completion December 2027; no promised publication date.

Patidegib Gorlin-syndrome study completes follow-up; results forecast for November

Source date: 2026-09-17. Evidence: Sponsor development milestone.

The phase 3 SGT-610 study has completed its final patient visit; the sponsor forecasts topline results in late November 2026. SGT-610-01 tests topical patidegib 2% versus vehicle for 12 months to prevent new facial basal-cell carcinomas in adults with Gorlin syndrome. It enrolled 113 people; 102 completed. Database lock and analysis are next, with assignments still blinded.

Limitations: No efficacy or comparative safety result is released. This inherited high-risk cancer-prevention setting does not establish cosmetic benefit or prevention in the general population.

Safety: The completed-visit announcement contains no new outcome-level safety analysis.

U.S. status: Investigational; trial completion and designations are not approval.

Next milestone: Late November 2026 topline: sponsor forecast, not a scheduled day.

Sol-Gel, September 17.

Primary topic: Cancer treatment, vaccines and recurrence prevention. Stable event key: sgt610_2026-09-17_last_visit.

Ketamir-2 prepares a controlled neuropathy trial

Source date: 2026-09-17. Evidence: Sponsor protocol-development announcement.

MIRA submitted a phase 2a chemotherapy-induced neuropathy protocol for ethics review; site initiation is targeted for Q1 2027, not confirmed underway. After FDA feedback, the planned randomized double-blind crossover study compares oral Ketamir-2 300 mg, 600 mg and placebo across three seven-day periods with washouts in adults with persistent moderate-to-severe chemotherapy-induced peripheral neuropathy. Enrollment size and a matched registry identifier were not established in the release.

Limitations: Healthy-volunteer phase 1 tolerability does not establish pain relief, function or chronic safety in patients. This oral NMDA-modulator program is distinct from approved suzetrigine and from psychedelic depression trials.

Safety: Sponsor reports no serious events or dose-limiting toxicity in phase 1; phase 2 comparative safety and CNS effects remain unmeasured.

U.S. status: Investigational; FDA protocol review and an active IND are not marketing approval.

Next milestone: Q1 2027 site initiation is conditional on IRB review and activation. An October 5–6 presentation concerns phase 1/development, not phase 2 efficacy.

Primary topic: Non-opioid pain treatments. Stable event key: ketamir2_2026-09-17_phase2_protocol.

Finerenone gains U.S. approval for CKD associated with type 1 diabetes

Source date: 2026-09-17. Evidence: Regulatory action; September 2026 U.S. prescribing information verified.

The formerly pending type 1 diabetes indication is now approved. The label specifically describes UACR reduction expected to reduce sustained eGFR decline and end-stage kidney disease. FINE-ONE randomized 242 adults and measured albuminuria over six months. The new label infers kidney-event benefit from type 2 diabetes outcome trials together with pharmacodynamic and safety evidence in type 1 diabetes. The approval expands the indication; it does not represent a new trial directly demonstrating dialysis prevention.

Limitations: Bayer announced the approval on September 17; the dated FDA action letter was not retrieved. The indication should not be generalized to all non-diabetic CKD.

Safety: Hyperkalemia remains the principal monitoring issue; strong CYP3A4 inhibitors and adrenal insufficiency are contraindications. Pooled 14% versus 6.9% hyperkalemia figures in the label belong to type 2 diabetes trials, not FINE-ONE.

U.S. status: Approved in the U.S. for adults with CKD associated with type 1 diabetes.

Next milestone: Implementation and longer-term evidence; no firm date for the next readout was verified.

Primary topic: Kidney preservation and earlier detection. Stable event key: finerenone_2026-09-17_US-T1D-CKD-approval.

Deramiocel two-year follow-up scheduled at World Muscle Society

Source date: 2026-09-17. Evidence: Sponsor conference/development announcement.

Capricor will present HOPE-3 extension and delayed-start analyses in Duchenne muscular dystrophy. Of 106 participants originally randomized, 82 reached the 24-month timepoint: 40 originally assigned to deramiocel and 42 originally assigned to placebo. WMS runs September 29–October 3, with the oral presentation scheduled for October 3. The second year is open-label, with delayed-start and natural-history analyses.

Limitations: The notice provides no numerical 24-month effect or safety estimates. The extension does not maintain a randomized placebo comparison or establish general muscle-preservation efficacy. Earlier 12-month results are baseline evidence, not new September 17 efficacy findings.

Safety: New long-term safety tables pending.

U.S. status: Investigational. The sponsor lists an FDA target action date of November 22, 2026; this does not guarantee approval.

Next milestone: WMS, September 29–October 3, with the oral presentation on October 3. FDA target action date: November 22, 2026.

Primary topic: Muscle building and preservation. Stable event key: deramiocel_2026-09-17_WMS-schedule.

Safety detail added to already recorded AgelessRx pilot

Source date: September 2 pilot; safety detail checked September 18. Evidence: Evidence correction to existing pilot.

The current sponsor page reports a serious adverse event omitted from the notebook summary. It does not establish that a new safety event occurred on September 17. The same 90-day pilot enrolled 26 participants and analyzed 18. Seven of 26 participants experienced adverse events, and three withdrew. One myocardial infarction occurred in the multi-therapy arm and was assessed as possibly associated with physical activity. The page now says it was last updated on September 17; earlier findings were first recorded as September 2.

Limitations: Bundled intervention, small groups, sponsor-only reporting and attrition; no causal attribution to any drug established. Do not equate unchanged laboratory panels with an absence of adverse events.

Safety: The myocardial infarction was a serious adverse event. Mixed cognitive and maximal oxygen-uptake results should also remain visible.

U.S. status: No approval of a longevity drug or regimen is established.

Next milestone: The sponsor plans a one-year trial with 186 participants; registration and actual start were not confirmed.

Resistance-training study during tirzepatide treatment is listed as recruiting

Source date: 2026-09-17. Evidence: Trial registry; prospective study to follow.

A randomized study will measure whether resistance training preserves contractile thigh muscle during tirzepatide treatment. The Technical University of Munich study, NCT07777575, plans to enroll 150 adults with obesity and compare 24 weeks of resistance training with usual care. MRI endpoint assessment is blinded. The primary endpoint is change in contractile thigh muscle volume. The record was first posted on August 20 and updated on September 17; the trial is listed as recruiting.

Limitations: September 7 start is still estimated in the primary registry despite mirrors calling it actual. Muscle volume is distinct from strength and everyday function; no results available. This is a training adjunct study, not a new muscle drug.

Safety: Prospective safety data pending.

U.S. status: Approved tirzepatide is the background treatment; no new indication from registration.

Next milestone: Estimated primary/overall completion May 31, 2027, not a promised readout date.

NCT07777575 current primary registry.

Primary topic: Muscle building and preservation. Stable event key: tirzepatide-resistance-training_2026-09-17_registry.

New study targets emotional blunting during antidepressant treatment

Source date: 2026-09-17. Evidence: new trial registration.

A newly registered phase 4 study will examine adjunctive lumateperone for emotional blunting in MDD. NCT07824206; Future Options Research with J&J collaboration, planned 30 adults, open-label single group. Eight-week Oxford Depression Questionnaire change is the primary endpoint. Registry status not yet recruiting; September 2026 start remains estimated.

Limitations: No results or comparator. Emotional blunting can reflect illness and treatment; the design cannot isolate a drug-causal explanation or comparative benefit.

Safety: No study safety results yet. Phase 4 designation does not establish benefit for this specific symptom.

U.S. status: Lumateperone has an adjunctive MDD indication, but this specific emotional-blunting claim is being studied.

Next milestone: Registry estimate: Primary completion January 2028; overall completion March 2028. Not promised publication dates.

KetSet will compare the therapeutic framing of ketamine

Source date: 2026-09-17. Evidence: new randomized study registration.

KetSet will compare psychedelic versus biomedical framing while keeping ketamine treatment the same. NCT07824973, Sweden; planned 120 adults with TRD, randomized parallel arms, participants/staff unblinded and outcome assessors masked. Primary endpoint MADRS change at week 12. Both groups receive six infusions; support/context differs. Not yet recruiting, September start estimated.

Limitations: No results. It tests context, not ketamine versus placebo or psilocybin. Registry title says open-label while design clarifies assessor masking.

Safety: Adverse events, discontinuation, suicidality and longer-term hospital outcomes are planned, not demonstrated.

U.S. status: No U.S. approval change; intravenous ketamine depression use remains distinct from intranasal esketamine approval.

Next milestone: Registry estimate: Primary completion December 2029; overall completion August 2030.

Small sponsor studies explore skin care after medication-associated weight loss

Source date: 2026-09-17. Evidence: sponsor conference report; preliminary human studies.

Allergan reports early skin-quality improvements in people taking weight-loss medicines, from two very small single-center studies. One 12-week skincare study had nine completers; a six-week DiamondGlow plus skincare study had 11 people with at least one post-treatment assessment. Selected investigator-rated dryness, texture, crepiness and other measures improved.

Limitations: The release supplies no randomized comparator, numerical effect sizes, uncertainty intervals or enrollment denominators. Its perceived younger-appearance claims are participant impressions, not biological-age reversal or evidence of substantial loose-skin correction.

Safety: Adverse-event counts were not supplied; do not describe the regimens as proven safe.

U.S. status: No new FDA indication or approval announced.

Next milestone: Posters at Science of Skin Summit September 17–20; full controlled replication and durability results undated.

Mosunetuzumab combination meets phase 3 lymphoma endpoint

Source date: 2026-09-16. Evidence: Sponsor topline; randomized phase 3.

CELESTIMO reports improved progression-free survival with mosunetuzumab plus lenalidomide in previously treated follicular lymphoma; overall survival is immature. NCT04712097 compares mosunetuzumab plus lenalidomide with rituximab plus lenalidomide after at least one prior treatment. The registry lists 478 actual participants. Genentech reports that the interim primary endpoint was met, without releasing a hazard ratio or absolute PFS estimates.

Limitations: PFS benefit must not be called a demonstrated survival benefit. The study supports a potential earlier-line indication and confirmatory regulatory review; neither action is completed by the topline announcement.

Safety: Sponsor reports no new safety signal, but comparative adverse-event tables are pending. Known mosunetuzumab risks include serious cytokine-release syndrome and infections.

U.S. status: Existing third-line-or-later monotherapy approval is separate from the investigational second-line combination.

Next milestone: Full results at an unnamed upcoming medical meeting and regulatory submissions; no firm date.

Small psilocybin subgroup study includes personality disorders

Source date: 2026-09-16. Evidence: peer-reviewed accepted article; exploratory secondary analysis.

A September 16 paper reports a ten-person TRD subgroup with co-occurring personality disorders, usually excluded from psychedelic trials. NCT05029466; nine had cluster B and one cluster C personality disorder. Three dropped out. Mean MADRS was 30.4 before treatment and 22.3 at six months; trajectories varied substantially. Registry parent trial enrolled 30 and completed in 2023.

Limitations: No controlled efficacy estimate for this subgroup; tiny selected sample, secondary analysis and attrition. Abstract reviewed; accepted-manuscript PDF access failed. Do not describe this as proven personality-disorder treatment or suicide prevention.

Safety: No serious adverse events reported in abstract; inadequate size to establish psychiatric safety or exclude uncommon harm.

U.S. status: Psilocybin remains investigational for these indications.

Next milestone: No announced date: Larger controlled studies and complete safety reporting needed.

PST-611 geographic-atrophy presentation scheduled for October 2

Source date: 2026-09-16. Evidence: announced conference presentation; recovered phase 1 baseline.

PulseSight announced an October 2 EURETINA presentation. The six-person phase 1 findings substantially repeat its May ARVO report, rather than constituting a fresh efficacy trial. PST-611-CT1 / NCT07024732 studies nonviral transferrin-gene delivery. Sponsor treated analysis: six patients, two dose levels, 16 weeks. May report says BCVA remained stable and cites exploratory anatomical/patient-reported signals. Registry lists eight enrolled and completion April 7; enrollment is not necessarily the treated analysis denominator.

Limitations: Uncontrolled, safety-focused study with no quantified comparative efficacy. September release also says twelve-month study in its boilerplate despite four-month follow-up in the body; do not infer validated twelve-month benefit.

Safety: May report: predominantly mild ocular AEs, two moderate events, no treatment-emergent SAE or intraocular inflammation reported.

U.S. status: Investigational, not U.S.-approved.

Next milestone: Scheduled conference presentation: EURETINA Session 6 #138, 13:05 in Vienna. May sponsor forecast: phase 2a start H2 2026 and results 2028, contingent on authorization; actual start not verified.

Dual-target RNAi lowers LDL, ApoB and triglyceride biomarkers

Source date: 2026-09-15. Evidence: Sponsor interim topline.

Arrowhead reports early ARO-DIMER-PA human data targeting PCSK9 and APOC3 with one RNAi molecule. The placebo-controlled phase 1/2a dose-escalation study, NCT07223658, plans to enroll up to 78 adults with mixed hyperlipidemia. Reported mean maximal reductions after a single dose were 54% for LDL-C, 50% for ApoB, 73% for triglycerides and 61% for non-HDL cholesterol. Target-protein reductions were 72% for PCSK9 and 88% for APOC3.

Limitations: Analyzed cohort sizes, uncertainty intervals, placebo-adjusted estimates and common measurement timepoints were not provided. Maximal biomarker reductions do not establish cardiovascular benefit and cannot support a comparable endpoint plot without further details.

Safety: Injection-site events and headache were the most common adverse events. No drug-related serious adverse events were reported. Safety with multiple doses remains under study.

U.S. status: Investigational; no U.S. approval.

Next milestone: Additional congress details are undated. The registry lists the trial as recruiting, with estimated primary completion on September 30, 2026 and overall completion in July 2027. These are not promised results dates.

SPLENDID improves cross-ancestry polygenic prediction in biobank analyses

Source date: 2026-09-14. Evidence: Peer-reviewed human-genetic methods study.

SPLENDID models ancestry as a continuum and improves prediction in diverse biobank populations; clinical benefit from using it has not been tested. The study combined simulations with All of Us (224,364 people) and UK Biobank (340,140). It reports improved prediction, particularly for non-European and admixed ancestries, without requiring discrete ancestry labels.

Limitations: This is prediction/model validation, not a randomized test of risk-guided prevention, and not validation of embryo ranking or long-term offspring benefit. The original abstract was retrieved through Europe PMC; full model-performance tables were not appraised.

Safety: Clinical harms and downstream management effects were not evaluated.

U.S. status: Publication does not establish a newly approved U.S. screening test.

Non-incretin CRB-913 reports phase 1b weight-loss data

Source date: 2026-09-14. Evidence: Sponsor topline.

The oral peripheral CB1 inverse agonist adds a non-incretin obesity program to the broader metabolic watch. CANYON-1, NCT07310901, randomized 254 U.S. adults with obesity without diabetes. After 12 weeks of treatment, efficacy-estimand weight loss was 2.8%, 3.3% and 5.0% across active doses versus 0.0% with placebo (all p < .0001). The highest-dose arm included 62 participants; placebo included 66. Four weeks of follow-up followed treatment. The registry retains an older May 29 snapshot, with estimated enrollment of 252 and no posted results; preserve the later sponsor-reported analysis population.

Limitations: Short phase 1b study, no uncertainty intervals or long-term outcomes; sponsor GLP-1 comparisons are cross-trial, not head-to-head. Do not label CRB-913 an incretin or add it to hormone-mimetic plots without explicit scope change.

Safety: Adverse-event discontinuations ranged from 3.1% to 13.1%; irritability affected 6.2%–9.7% across active doses versus 0% with placebo. No serious/severe psychiatric event or suicidality reported. Gastrointestinal symptoms occurred; long-term neuropsychiatric safety remains unestablished.

U.S. status: Investigational.

Next milestone: A detailed presentation is scheduled for ObesityWeek, November 14–17, 2026. The sponsor targets a phase 2 start in the first half of 2027.

IVIG add-on improves early inflammatory-myopathy disease activity

Source date: 2026-09-14. Evidence: Peer-reviewed randomized human trial.

TIME IS MUSCLE supports earlier IVIG add-on in a small newly diagnosed inflammatory-myopathy population. The double-blind trial randomized 44 adults, all treated with high-dose prednisone. The reported primary-outcome sample included 42 participants: 23 receiving IVIG and 19 receiving placebo. At week 12, Total Improvement Score was 60.0 (95% CI 52.6–67.4) versus 42.5 (95% CI 30.6–54.4), p = .01. Moderate improvement occurred in 91% versus 53%. Trial identifier: EudraCT2020-001710-37-NL.

Limitations: This was a single-center study with short follow-up and a disease-specific composite endpoint. It does not establish healthy muscle gain or muscle preservation during GLP-1 treatment. The major-response analysis was post hoc. Full supplemental analyses were not reviewed.

Safety: One asymptomatic deep-vein thrombosis occurred in the IVIG group.

U.S. status: No new U.S. label action established by this publication.

Next milestone: Further comparative evidence and longer follow-up are undated.

Photothermal PRP does not outperform conventional PRP on melasma severity

Source date: 2026-09-12. Evidence: peer-reviewed randomized human study; original abstract reviewed.

A small split-face trial found no superiority on clinician-rated melasma severity despite a difference in instrument-measured pigmentation. TCTR20260318004; 26 women received three monthly sessions, with photothermal-activated and conventional PRP on opposite sides. At 12 weeks both mMASI scores improved; no between-treatment difference at any assessment. Photothermal PRP had lower melanin index at week 12.

Limitations: No untreated/sham side, short follow-up, small female-only sample. No numerical between-group effect or confidence interval in abstract; nonsignificance is not proof of equivalence. Publisher contents page verifies first publication September 12.

Safety: Mild injection-site bruising was the only reported adverse event; sample cannot resolve uncommon harms.

Silica-enriched toothpaste improves sensitivity but not measured mineral gain

Source date: 2026-09-12. Evidence: peer-reviewed pilot randomized human study; abstract reviewed.

A pediatric enamel-defect trial found a pain-sensitivity signal but no superiority over fluoride toothpaste on its mineral-gain endpoint. RBR-6d3g5hp / U1111-1327-7119; triple-blind pilot, 30 children aged 6–12, 95 teeth, 45 days. REFIX Booster versus 1450 ppm fluoride. Between-group change in fluorescence ΔF: −0.43 percentage points, 95% CI −2.73 to 1.86, p=.711; ΔFmax also nonsignificant. Sensitivity improved more, p=.011. Models accounted for teeth clustered within children.

Limitations: Short pilot; fluorescence is a surrogate, not replacement of missing enamel or tooth regeneration. No relevant aesthetic difference; secondary pain signal needs replication.

Safety: No adverse effects reported in this small short study.

U.S. status: No new U.S. authorization or availability established.

Next milestone: Definitive trial and durability follow-up undated.

Advertising review challenges precision claims for embryo selection

Source date: 2026-09-11. Evidence: Advertising self-regulatory decision; appeal announced.

NAD recommended changing or discontinuing specific Nucleus IVF+ embryo-comparison claims; Nucleus announced an appeal. The decision questioned whether submitted evidence validated ranking a group of sibling embryos and the advertised numerical differences in disease risk, height and IQ, as well as definite eye/hair-color predictions.

Limitations: This is an advertising self-regulatory finding, not an FDA ban, a new clinical trial, or a determination that all genetic testing lacks value. Keep it separate from inherited-disease PGT-M and the non-invasive PGT-A trial.

Safety: No new clinical safety outcome was measured.

U.S. status: Commercial offering and claim substantiation are separate; no change in FDA authorization is established.

Next milestone: Appeal outcome; no date announced.

NCT07214311 is a recruiting randomized placebo-controlled phase 2/3 study targeting 240 participants. It measures treatment failure through 12 months after debridement, antibiotics and implant retention, with surgery, survival and function also tracked. The registry lists a February 3, 2026 global start; September is a site-enrollment report, not global trial initiation.

Limitations: This newly added formal antimicrobial program addresses biofilm-associated infection, not cartilage regeneration. Laboratory activity and earlier small studies do not establish superiority, broad resistant-infection efficacy or implant retention benefit.

Safety: No new comparative efficacy or safety results were reported.

U.S. status: Investigational; Orphan/Fast Track/QIDP designations are not approval.

Early BEST1 gene-therapy findings include a corrected visual-acuity chart

Source date: September 9 announcement; corrected September 11. Evidence: sponsor phase 1/2 topline and SEC correction.

Opus reported early visual-test signals in five BIRD-1 participants on September 9 and corrected the visual-acuity conversion on September 11. NCT07185256; three BVMD participants had three-month follow-up, two ARB participants six months. The company reported gains on at least one visual test per participant; denominators vary by measure. September 11 correction gives treated-eye three-month ETDRS changes +12, +1, −10, +2 and +6 letters, versus fellow-eye +4, −7, −8, −5 and +2. Use revised presentation, not the original mean chart.

Limitations: Open-label n=5, multiple outcomes and mixed follow-up; fellow eyes are not a randomized control. Restricted to inherited BEST1 retinal disease, not common AMD. Registry enrollment estimate 10 predates sponsor cohort expansion.

Safety: No serious adverse events, dose-limiting toxicities or intraocular inflammation reported; related AEs mild/moderate. Long-term and rare risks unresolved.

U.S. status: Investigational; reported FDA endpoint discussions are not approval.

Next milestone: Scheduled presentation / sponsor forecast: EURETINA October 4, 2026, 13:15–13:21 CEST; cohort 2 dosing completion Q4 2026, further data Q2 2027 and potential pivotal dosing in 2027.

Platelet concentrates improve early extraction healing and selected bone dimensions

Source date: 2026-09-11. Evidence: peer-reviewed randomized human study; accepted early article.

A 60-person extraction trial found early healing and imaging gains with blood-derived concentrates; it did not study new teeth or long-term tooth retention. TCTR20251028003, retrospectively registered. Impacted lower-third-molar extraction; 20 each advanced platelet-rich fibrin, concentrated growth factors, natural healing. Both interventions improved early pain/swelling/healing. At 3 months A-PRF buccal-wall height change +1.32 ± 0.72 mm versus +0.01 ± 1.6 mm with CGF and −0.07 ± 1.12 mm controls; both preserved width.

Limitations: One small extraction setting; accepted version may be edited. Three-month radiographic dimensions do not establish durable functional regeneration or superior implant survival.

Safety: Accessible abstract does not provide a complete adverse-event table.

U.S. status: No new FDA authorization or generalized dental-regeneration claim.

Next milestone: Longer comparative follow-up undated.

Rapamycin pilot reports brain-perfusion increases in APOE4 carriers

Source date: 2026-09-10. Evidence: Peer-reviewed single-arm human pilot.

A four-week study reports a cerebral blood-flow signal in cognitively normal APOE4 carriers; it did not test prevention of dementia or longer life. Twenty-three adults aged 45–65 completed the study: nine APOE4 carriers and 14 noncarriers. They received rapamycin 1 mg/day for four weeks. The primary cerebral blood-flow endpoint increased by more than 15% in multiple regions among carriers; noncarriers had no significant change. Secondary metabolic, inflammatory and microbiome measures were explored. DOI: 10.1177/0271678X261490342.

Limitations: There was no placebo or control treatment. Genotype groups were tiny, follow-up lasted four weeks, and multiple exploratory measures were examined. Appraisal was limited to the abstract; detailed safety tables and the registry record were not retrieved.

Safety: The abstract describes minimal adverse effects but provides no event counts; long-term safety cannot be inferred.

U.S. status: Rapamycin is not approved for longevity or Alzheimer’s prevention.

Next milestone: No date for a subsequent trial or readout was verified.

HERMES trial is terminated

Source date: 2026-09-10. Evidence: Trial-registry development update.

Fresh registry confirms ziltivekimab HERMES termination, superseding the earlier continuation forecast. NCT05636176 lists 4,899 actual participants, actual completion September 3 and last update September 10. No results posted. This is a heart-failure program followed as context to the broader ziltivekimab prevention program.

Limitations: The reason field says only that the trial is no longer looking for participants. The registry alone does not establish futility or a safety cause. ATHENA, NCT06200207, remains active and not recruiting in an older September 1 record, so later reports that it stopped remain unreconciled.

Safety: No new safety or outcome estimates posted.

U.S. status: Investigational.

Next milestone: Detailed program explanation and full ZEUS results undated.

ALZ-101 phase 2 enters operational preparation

Source date: 2026-09-10. Evidence: sponsor trial-development announcement.

Alzinova announced operational start-up of its therapeutic Alzheimer vaccine phase 2 study, not first enrollment or dosing. Global randomized placebo-controlled study plans about 120 early-AD participants, ADCOMS change at 52 weeks as primary endpoint and 92-week total follow-up. CRO kick-off completed; regulatory/ethics submissions and site preparations ongoing.

Limitations: Preparation is not recruiting status or efficacy evidence. No matched phase 2 registry ID verified.

Safety: No new safety result in announcement.

U.S. status: Investigational therapeutic vaccine, not approved prevention.

Next milestone: Sponsor forecast: First enrollment around the turn of 2026/2027; no results date announced.

Inhaled RNA interference reaches human lung target-engagement evidence

Source date: 2026-09-09. Evidence: Peer-reviewed randomized phase 1/2a study.

ARO-RAGE reduced its lung inflammatory target in early human research; fewer asthma attacks or COPD benefit have not been demonstrated. NCT05276570 studied healthy volunteers and mild-to-moderate type-2 asthma. The published cohorts include 58 treated healthy volunteers and 19 treated asthma patients, with 29 placebo participants. The study focused on safety and target engagement; pulmonary-function measures were safety assessments, not powered efficacy tests.

Limitations: The registry lists 127 overall participants, while the published cohorts total 106; the abstract active-treatment counts must not be represented as all randomized participants. No clinical-outcome benefit is inferred from soluble-RAGE reduction.

Safety: Headache and respiratory infections were common. COVID-19 occurred in 14/58 treated healthy volunteers versus 2/23 placebo. One delayed pre-eclampsia serious event was assessed unrelated; no asthma serious events. Small cohorts cannot settle uncommon harms.

U.S. status: Investigational inhaled siRNA; no U.S. approval.

Next milestone: Separate phase 2a NCT07241546 is recruiting (target 36); primary completion December 2027 is a registry estimate, not a promised readout.

COMP005 reports one-year follow-up with additional psilocybin doses

Source date: 2026-09-09. Evidence: sponsor topline open-label extension.

COMP005 Part C reported improvement through week 52 after permitted additional COMP360 dosing. Original trial randomized 258 people with TRD. Approximately 70% entered open-label weeks 26–52; 90 originally assigned 25 mg received a second or third dose and 52 originally assigned placebo received their first 25 mg dose. Sponsor reports a 13-point mean baseline-to-week-52 MADRS reduction in the original active-arm repeat-dose group, with 40–45% response and about 30% remission over six weeks after Part C dosing.

Limitations: Open-label, selected continuation, attrition and redosing. This is not randomized evidence that one dose lasts a year; confidence intervals and complete denominators at each time point not supplied.

Safety: No new safety findings reported; detailed rates absent.

U.S. status: Investigational; rolling filing is not approval.

Next milestone: Sponsor forecast: Rolling NDA completion Q4 2026; conditional U.S. launch H1 2027, subject to approval/rescheduling.

Non-invasive embryo chromosome screening does not improve the primary pregnancy outcome

Source date: 2026-09-08. Evidence: Peer-reviewed randomized trial; accepted early article.

A large randomized trial found no significant improvement in ongoing pregnancy or live birth after the first transfer with non-invasive PGT-A versus morphology-based selection. NCT04339166 enrolled women aged 35–42 with at least two blastocysts at 13 Chinese centers. The modified intention-to-treat analysis included 1,152 couples: ongoing pregnancy beyond 12 weeks was 38.7% (221/571) versus 36.8% (214/581), adjusted p=.49; live birth was 37.0% versus 35.5%, p=.59. Early miscarriage was lower in a secondary analysis.

Limitations: This tests cell-free DNA from embryo culture medium, not embryo-biopsy PGT-A or polygenic PGT-P. First-transfer success is not cumulative live birth per retrieval. The abstract lists 1,189 allocated and 1,152 analyzed; these denominators are kept distinct.

Safety: No serious adverse events reported; maternal/neonatal differences were not significant. Long-term offspring outcomes were not established.

U.S. status: Research result, not a new U.S. authorization or proof of routine clinical utility.

Next milestone: Final publication and further clinical-utility evidence; no dependable next date.

Alzheimer Tau Platform adds combination-treatment research

Source date: 2026-09-08. Evidence: recovered baseline; current trial registry.

The Alzheimer Tau Platform is an existing recruiting phase 2 program studying tau therapies alone or with donanemab. NCT06957418, USC; master-protocol planned enrollment 900, actual start July 6, 2026. Adults 50–80 with late preclinical/early prodromal AD. Launch regimen AADvac1; later Tau2 regimen is unnamed. Randomized masked comparisons include tau therapy alone, combination and donanemab alone; primary outcome tau PET reduction.

Limitations: Master-protocol target is not enrollment for each arm. Biomarker primary outcome is not established cognitive benefit, and no results are posted.

Safety: No new safety data; combination safety remains under study.

U.S. status: Investigational regimen; donanemab approval does not authorize this preventive/combination use.

Next milestone: Registry estimate: Primary/overall completion August 31, 2028; no announced publication date.

Higher-dose vitamin D does not prevent fractures in a largely sufficient population

Source date: 2026-09-07. Evidence: Peer-reviewed randomized trial; primary abstract reviewed.

The Finnish FIND trial found no fracture reduction from 1,600 or 3,200 IU/day in older adults who were largely vitamin-D sufficient. NCT01463813 randomized 2,495 adults to the two doses or placebo. Over mean 4.1 years of supplementation, fractures numbered 73, 52 and 49 respectively: HR 1.52 (95% CI 1.06–2.18) for 1,600 IU and 1.07 (0.72–1.58) for 3,200 IU versus placebo. Extended follow-up also showed no benefit.

Limitations: Baseline vitamin D was measured in a 550-person subcohort (mean 74.8 nmol/L). This does not answer treatment of established deficiency or all high-risk osteoporosis populations. No simple dose-response explanation is established by the lower-dose imbalance.

Safety: The increased fracture rate in the 1,600-IU group is retained as a safety-relevant finding; fracture subtypes did not differ significantly. Detailed non-fracture adverse events were not available in the abstract.

U.S. status: A research result, not a new drug approval or individualized supplementation instruction.

Next milestone: Full-paper appraisal and replication; no dated next milestone.

Previously inaccessible melasma pilot now appraised from indexed original text

Source date: 2026-09-07. Evidence: peer-reviewed pilot randomized human study; gap resolution.

A multicomponent cosmetic cream and triple-combination comparator both improved short-term melasma scores; equivalence was not established. Singapore single-blind split-face pilot, 30 enrolled, 56 days. Cream includes tomato extract, cysteamine, niacinamide and other ingredients. Mean mMASI changes were −1.471 with cosmetic cream and −1.394 with tretinoin/hydroquinone/fluocinolone comparator.

Limitations: Small feasibility-based sample; not powered for noninferiority, different application schedules, no post-treatment relapse follow-up. Cannot attribute the effect to tomato extract alone. Direct page is under maintenance; indexed primary methods/abstract/discussion were readable.

Safety: Mild self-limited adverse events reported, 6.7% per arm; does not establish long-term safety.

U.S. status: Cosmetic formulation study, not a new FDA-approved melasma indication.

Next milestone: Confirmatory controlled study and post-treatment follow-up undated.

Previously inaccessible metformin biomarker trial recovered

Source date: 2026-09-03. Evidence: Historical evidence recovered.

The September 3 paper resolves a prior access gap but measures cellular biomarkers rather than healthspan. The trial randomized 127 North Indian adults aged 30–60 with prediabetes; 112 completed 24 weeks. Leukocyte telomere elongation was 29.6% with metformin versus 4.4% with placebo (between-group p < .0001). Trial identifier: NCT07302932.

Limitations: No survival, disability or disease-incidence endpoint; marked biomarker response needs replication. Registry first posted December 24, 2025, after actual January 30, 2025 completion: retrospective registration. Registry reports 112 enrolled, whereas the publication reports 127 randomized and 112 completers. Preserve those distinctions. December 2026 issue date is not original publication date.

Safety: The publisher reports early mild gastrointestinal symptoms in eight participants (14.3%) receiving metformin and two (3.6%) receiving placebo. These resolved without discontinuation, and no serious adverse events occurred. The percentages use completers rather than randomized participants as denominators.

U.S. status: No longevity indication.

Older missing ZEUS negative outcome and infection signal

Source date: 2026-07-31. Evidence: Historical evidence recovered.

Ziltivekimab lowered inflammation but did not lower MACE in ZEUS. ZEUS enrolled more than 6,300 participants with established atherosclerotic cardiovascular disease, CKD and hsCRP of at least 2 mg/L. The hazard ratio for three-component MACE was 0.99 (95% CI 0.88–1.11).

Limitations: The July 31 announcement was omitted from the baseline; it is not a new September trial result. Numerical infection counts remain unavailable. The failure should not be generalized to every anti-inflammatory target.

Safety: Overall adverse-event and serious-adverse-event rates were similar between groups, but serious infections were more frequent with ziltivekimab. No difference in all-cause mortality was observed.

U.S. status: Investigational.

Next milestone: Full results are expected at an unspecified 2026 scientific meeting. The July forecast that HERMES would continue must be updated against its termination record.
