## Current state

A new décolletage-wrinkle publication reports a randomized SB-NASHA benefit at 12 weeks, with common injection-site reactions. The safety population is broader than the randomized efficacy cohort. Wrinkle response remains distinct from generalized skin regeneration, durable lifting or volume restoration.

Status: active. Selective initial baseline completed September 12, 2026. Human research worldwide; U.S. status identified separately.

## Questions to follow

- Which therapies produce visible, durable improvements in wrinkles, crepey skin and laxity, including changes after weight loss?
- Can structural-protein replacement improve skin beyond existing topical treatments, injectables and devices?
- How much tightening is achieved, for which body sites and degree of laxity, with what downtime and adverse effects?
- Track drugs, devices and procedures. Distinguish measured collagen/elastin expression from appearance, and modest skin-quality gains from correction of substantial excess skin. Cross-link [facial volume](22_facial_volume.md), [scars](21_scars.md) and [localized fat](06_localized_fat.md).

#### Hyaluronic-acid microinjections improve chest wrinkles in a randomized trial

An October 9 peer-reviewed publication reports SB-NASHA (Restylane Skinboosters Vital Lidocaine) microinjections for moderate-to-severe décolletage wrinkles. The main study randomized 192 women, 144 to three treatment sessions and 48 to no treatment; 19 additional women with Fitzpatrick V–VI skin entered safety follow-up without randomization.

At week 12, blinded-evaluator response (at least one wrinkle-severity grade) was 73.6% versus 18.8%. The [posted registry](https://clinicaltrials.gov/study/NCT04717934) reports a 54.9-percentage-point difference (95% CI 41.7–68.0). At week 32, observed-case response was 80.8% versus 40.9%; later percentages have different evaluable denominators.

**Safety:** Treatment-related adverse events affected 32/163 treated participants (19.6%); none were serious. Diary-reported injection reactions were more common: 92.4% after the first session, usually resolving within one to two weeks. Some adverse events lasted longer, so brief downtime is not universal.

**Limits and U.S. status:** This Galderma-funded study had no sham injections; randomized efficacy largely concerns skin types I–IV. Safety follow-up extended to week 56 but cannot exclude rare harms. No current U.S. approval for this precise formulation/indication was verified. Registry results first posted July 15; October 9 is publication, not first disclosure.

#### HU-045 glabellar-line comparison enters the Phase3 registry

**First registry posting October 13, 2026; reported actual start September 3.** NCT07870694, Huons Biopharma, lists a recruiting randomized, participant/investigator-masked Phase 3 comparison in 390 planned adults aged 19–65 with moderate/severe glabellar lines. HU-045 and Xeomin are given at five 4-unit glabellar sites. The primary measure at week 4 requires investigator-rated lines to become none/mild with at least two grades of improvement; later assessments and satisfaction extend through week 16.

**Limits/safety:** No efficacy or adverse-event results. Temporary neuromuscular wrinkle smoothing does not establish skin regeneration, laxity treatment or facial-volume restoration. The structured phase and official title say Phase 3, while the brief summary still says PhaseII; retain the metadata discrepancy pending clarification. Registry description associates HU-045 with botulinum toxinA/incobotulinumtoxinA, but does not establish interchangeability with Xeomin or superiority.

**U.S.:** This Korean study establishes no U.S. approval or availability for HU-045. **Next:** primary completion May 2027 and final completion August 2027 are registry estimates, not readout promises.

[Original registry](https://clinicaltrials.gov/study/NCT07870694).

**Remaining gaps:** Read canonical README, HOW_THIS_WORKS, WATCHLIST, catalog, state, LATEST, MILESTONES, LANDSCAPE and assigned current topic. September 5–October 13 window preserves seven-day overlap from September 12 full successful check. Retrieved every page of bounded public registry discovery (70 returned records; incidental matches excluded), and all named records. Full successful-check date must remain unchanged.; Europe PMC retrieval now succeeded, closing prior HTTP503 access failure. Initial broad query downloaded up to 100 per topic with many incidental/nonhuman matches; title/abstract MED retry retrieved hair 97/97, skin 27/27, scars 25/25, dental 100/147. Retrieved records are not an exhaustive worldwide full-paper/safety appraisal.; New HU-045 glabellar-line Phase 3 registry appraised. KB304 remains active/not recruiting with stale 2025 estimate; ellacor nine-person cohort remains completed/no results. Existing post-weight-loss and secretome/energy-device claims deduplicated. RLS-1496 hair plan belongs to hair_loss; October 12 AK investigator interview is newly posted secondary context, not verified wrinkle rejuvenation. Original EADV228 abstract, full photoaging numerical tables, device clearance/rare-harm census remain gaps.

**Informal-source coverage:** September 22 Hot Water episode with Christie Prendergast found in original Apple listing and secondary Podcast Rex description; secondary summary attributes a clinic secretome trial plan but full transcript fetch failed, original audio/time passage unverified and possible overlap with saved Acorn program unresolved. Kept as follow-up lead, not newly verified trial or quoted statement. October 12 RLS posts frame senescence changes as age reversal; this interpretation is not proven clinical rejuvenation. #### Recovered baseline: ultrasound laxity series has mixed objective and subjective outcomes

**Historical publication:** June 1, 2026; October 5 sponsor promotion is not a fresh efficacy readout. **Baseline addition.**

A prospective uncontrolled Hong Kong series treated 42 Asian adults aged 35–67 once with high-intensity, nonfocused parallel ultrasound across face, neck and submental regions. Physician-rated submental/neck improvement occurred in 83.3% at three months. The objective jawline/submental volume change was −1.04 ± 3.49 mL, p=.06; this did not establish a statistically reliable effect or selective fat reduction. The abstract's wrinkle/elastosis measure differs from the sponsor's headline laxity percentages; do not combine them into one durability response rate.

**Safety and limits:** Initial erythema affected 50% and edema 7.2%, resolving at follow-up; no other events were documented. Small size, no comparator, open assessments and missing full appraisal limit efficacy, durability and rare-harm conclusions. This is not correction of substantial post-weight-loss excess skin. Exact FDA-cleared indication/version should be read separately; no new U.S. clearance occurred in this report. **Next:** independent controlled comparisons and complete long-term data, undated.

[Original publication](https://onlinelibrary.wiley.com/doi/10.1002/lsm.70153); [October 5 sponsor release](https://sofwave.com/news/new-peer-reviewed-study-shows-sustained-improvement-in-skin-laxity-through-nine-months-after-a-single-sofwave-treatment-in-asian-patients).

**Remaining gaps:** Read the current assigned topic and canonical scope/state. Named original registry records refreshed; all pages of broad ClinicalTrials.gov discovery for September 5–October 9 were retrieved (41 records, with incidental matches excluded). This honors the seven-day overlap from the saved September 12 full successful check.; Europe PMC original-literature discovery again returned HTTP 503. Targeted primary publisher, sponsor and conference searches were performed, but the worldwide paper/regulator/procedure and original-audio census remains incomplete. Preserve last_successful_check_utc.; KB304 remains active/not recruiting with stale 2025 completion estimate; no Phase 2 actual initiation verified. Ellacor nine-person completed/no-results cohort and AnteAGE pilot registrations already captured October 6. FDA RF-microneedling communication remains older ongoing evaluation, not relabeled a new alert.; Sofwave June paper full text redirected to abstract; October 5 sponsor promotion deduplicated by actual paper date. September 22 multimodal RF RCT full appraisal and world device clearance/rare-complication census remain incomplete. PRX S acne-prone skin NCT07866131 is an observational cosmetic protocol, no outcome.

#### Small post-weight-loss micro-coring cohort is completed, without results

**Source date:** October 5, 2026 registry posting. **Reported actual completion:** August 21, 2026.

NCT07224880 now lists a completed prospective U.S. ellacor micro-coring cohort with nine actually enrolled adults aged 30–65. Its named GLP-1 cohort had lost at least 10% of baseline weight. Planned assessments cover safety and appearance through approximately 90 days across Fitzpatrick skin types I–VI. No results are posted. This small, uncontrolled cohort cannot establish durable correction of post-weight-loss laxity, broad skin-type safety or superiority to another procedure. Completion is a development milestone, not evidence of successful tightening or a new U.S. clearance. **Next:** results publication and longer controlled follow-up, with no verified release date.

#### Separate 12-person exosome/microneedling pilot disclosed after completion

**Source/event date:** September 30, 2026 first registry posting. **Historical study:** August–November 2025.

NCT07849205 discloses a completed, uncontrolled U.S. pilot in 12 adults aged 35–65 receiving mechanical microneedling with AnteAGE Exosomes, PEARL Multiplier and a multicomponent home-care regimen. Twelve-week outcomes include skin-retraction time, VISIA coarse-wrinkle area and water loss. No results are posted. This is a separate study from the already recorded 30-person biosome/radiofrequency comparison, not an independent replication of that regimen. Retrospective registration, no control and multiple concurrent products prevent attribution to exosomes. Planned elasticity/barrier measurements do not prove tissue regeneration or durable laxity correction; safety results and a new U.S. authorization are not established. **Next:** results and component-controlled replication, undated.

#### Biosome plus radiofrequency microneedling trial registered after completion

**Source/event date:** 2026-09-30. **Evidence:** New registry disclosure of a historical pilot; no results.

A newly posted registry record describes a completed, randomized, masked 30-person pilot of radiofrequency microneedling with AnteAGE Biosome Solution and accompanying skin care versus the same procedure with hyaluronic-acid glide and control skin care. Outcomes include early reactions, wrinkles/discoloration and adverse events through 60 days. Actual study dates were December 2024–June 2025; first posting was September 30, 2026. This is retrospective disclosure, not a newly recruiting trial or positive efficacy readout. The intervention uses lipid vesicles; it should not be described as a proven stem-cell or tissue-regeneration treatment. No results are posted and no U.S. approval for this regimen was verified. [Registry](https://clinicaltrials.gov/study/NCT07849556).

**Next milestone:** Results publication: date unknown.

#### Small split-face study tests polynucleotide add-on to radiofrequency microneedling

**Source/event date:** 2026-09-28. **Evidence:** Peer-reviewed primary abstract; full-text appraisal incomplete.

In a prospective study of 25 women, both facial sides received radiofrequency microneedling; the right side received 2 mL Rejuran polynucleotide and the left saline. At six weeks, pore-score improvement was 4.79 versus 0.95 and wrinkle-score improvement 5.04 versus 3.67 (both p<0.001). Dermal-thickness and elasticity differences were not significant. This fixed-side, small study does not establish durable tightening or regeneration, and score changes are not directly interpretable as percentage cosmetic improvement. Full adverse-event counts and longer follow-up were not available in the abstract; full text could not be appraised. This is not a U.S. approval update. #### IM-AGE registers a small study of less-invasive skin-aging measurements

**Source date:** 2026-09-22. **Evidence:** trial-registry registration.

A UK study will compare OCT imaging and skin-fluid biomarkers with biopsies while testing short exposures to marketed cosmetic products. ISRCTN10674073 targets 24 younger adults and adults aged 45+ with photodamage. Older participants receive four randomized product patches and control areas over 12 days; the primary focus is measurement validation. The September 22 record lists recruiting.

**Limitations:** No efficacy results. Biomarkers and exaggerated-use patches do not establish durable visible rejuvenation. The record labels registration prospective despite listing first enrollment September 14, before September 22 registration. Its plain-language September–December 2026 schedule also differs from structured completion May 3, 2027.

**Safety:** Planned biopsy and product-tolerability monitoring; no safety results.

**U.S. status:** No new U.S. treatment approval; this is a UK methods/cosmetic study.

**Next milestone:** Resolve registry chronology and completion discrepancies; public results timing unknown.

#### Hair-follicle secretome presentation announced, with very early skin-recovery data

**Source date:** 2026-09-23. **Event date:** 2026-09-23. **Evidence:** investigator/practice announcement; preliminary human study and conference schedule.

Bauman Medical announced an October presentation comparing a hair-follicle-derived secretome with PRP; its biochemical measurements do not establish better hair regrowth. The announcement describes a post-ablative-laser recovery pilot in 11 people, reporting 32–58% reductions in recovery symptoms versus comparators. It also compares proteins and exosome particles in preparations from the same donors. The cited NCT06477172 is a separate prospective observational study, recruiting with 500 planned participants and February 2027 estimated completion; its primary outcomes are short-term post-procedure symptoms and downtime.

**Limitations:** Sponsor/practice account without the complete abstract, allocation methods, confidence intervals or full clinical tables. Product-composition differences are not hair-density or durable rejuvenation outcomes. Investigator disclosed research and ownership relationships.

**Safety:** The release provides no detailed adverse-event table; small early recovery observations cannot establish rare or long-term safety.

**U.S. status:** The announcement explicitly says aHFS is investigational and not FDA cleared or approved.

**Next milestone:** ISHRS congress October 15–17, 2026; the release does not specify an exact presentation day. Await the actual abstract and controlled clinical outcomes.

#### IPL pilot finds no objective regimen advantage after multiplicity correction

**Source date:** 2026-09-15. **Event date:** 2026-09-15. **Evidence:** peer-reviewed randomized human pilot; abstract appraised.

A longer-pulse IPL regimen was more comfortable, but did not show a statistically reliable advantage on the measured objective skin outcomes. Twenty-seven Chinese participants with mild-to-moderate photoaging received three sessions: 80-ms/higher-fluence IPL (13) versus 15-ms/lower-fluence IPL (14). At three-month follow-up the pore-count difference was −264.5 (95% CI −466.9 to −62.1), nominal p=.0127, but Holm-adjusted p=.0635. The other four objective endpoints were also nonsignificant after correction. Pain and satisfaction favored 80 ms.

**Limitations:** Small exploratory trial; complete regimens differ in both pulse duration and fluence, so effects cannot be attributed to pulse width alone. No sham control. Full paper and registry matching remain gaps.

**Safety:** Mild self-limited adverse events; no serious events, blistering, edema, pigment alteration or scarring reported in this small sample.

**U.S. status:** No new U.S. clearance established.

**Next milestone:** Larger controlled comparisons and durability results undated.

Refreshed KB304 registry, which still has stale 2025 estimates, and searched sponsor updates. September 17 Allergan small skin-care studies already recorded and deduplicated.

**Remaining gaps:** September 22 multimodal RF/microneedle-RF/muscle-stimulation trial abstract lacks sample size and numerical effects; full paper not appraised, so no efficacy alert promoted. Allergan September 17–20 conference window has elapsed; no full poster/safety tables recovered. This does not prove presentation or positive completion. FDA RF safety communication was not freshly appraised in this run; rare-harm surveillance remains incomplete.

**Informal-source coverage:** Searched Reddit and podcasts. #### Small sponsor studies explore skin care after medication-associated weight loss

**Source date:** 2026-09-17. **Evidence:** sponsor conference report; preliminary human studies.

Allergan reports early skin-quality improvements in people taking weight-loss medicines, from two very small single-center studies. One 12-week skincare study had nine completers; a six-week DiamondGlow plus skincare study had 11 people with at least one post-treatment assessment. Selected investigator-rated dryness, texture, crepiness and other measures improved.

**Limitations:** The release supplies no randomized comparator, numerical effect sizes, uncertainty intervals or enrollment denominators. Its perceived younger-appearance claims are participant impressions, not biological-age reversal or evidence of substantial loose-skin correction.

**Safety:** Adverse-event counts were not supplied; do not describe the regimens as proven safe.

**U.S. status:** No new FDA indication or approval announced.

**Next milestone:** Posters at Science of Skin Summit September 17–20; full controlled replication and durability results undated.

[AbbVie original release](https://news.abbvie.com/2026-09-17-Allergan-Aesthetics-Presents-New-Clinical-Data-Evaluating-Skin-Quality-Improvements-in-Individuals-Taking-Medical-Weight-Loss-Medications).

**Coverage:** Searched human skin-quality, tightening, regenerative and device developments, including medical-weight-loss populations. Appraised Sep 17 AbbVie sponsor announcement. Refreshed original KB304 registry and FDA radiofrequency microneedling communication; no new FDA update was found on the existing safety notice. This is a focused review, not a complete re-review of every program.

**Remaining gaps:** Allergan full posters, original protocols, enrollment denominators and adverse-event tables were not available publicly in reviewed material. KB304 original registry has stale estimated completion dates and no results; 2027 Phase 2 sponsor guidance is not a confirmed start. NCT07661004 LED-mask/serum record was first posted June 22, not September; September estimated start remains unverified.

**Informal-source coverage:** Searched Reddit, podcast and talk leads. Reviewed Sep 8 Fill Me In episode description on injectables and regenerative combinations; no original trial readout was supplied and audio/transcript was not reviewed. Third-party summaries of skin-regeneration podcasts were not treated as clinical evidence.

#### KB304: collagen III and elastin gene delivery

Jeune Aesthetics/Krystal's investigational intradermal gene therapy is being developed for upper-chest wrinkles. **PEARL-2, [NCT06724900](https://clinicaltrials.gov/study/NCT06724900)**, randomized 19 women to KB304 or placebo; 18 were assessed after one participant withdrew before completing treatment. In the sponsor's July 24, 2025 report, investigator-rated improvement at three months occurred in 11/11 treated participants versus 2/7 controls; participant-rated improvement occurred in 8/11 versus 1/7. Reported adverse events were mild or moderate, with no severe or drug-related serious events. This is a small Phase 1 signal with short follow-up, not established long-term rejuvenation or evidence for other body sites. **Next:** the August 3, 2026 company update targets **Phase 2 initiation in 2027**, superseding the older first-half-2026 plan. This is a sponsor forecast for trial start, not a results or approval date. No U.S. marketing approval or dependable readout date was verified. #### Retinoids: the established comparison for photoaging

Topical tretinoin provides a useful efficacy benchmark for newer products. A two-year randomized vehicle-controlled study of **204 participants** found greater improvement in facial photodamage with tretinoin 0.05%, including wrinkles and uneven pigmentation. Increased procollagen staining was a separate biological measurement. The trial does not establish meaningful removal of loose skin after major weight loss. Monitor genuinely improved tolerability, adherence and comparative performance of newer formulations, rather than treating another collagen claim as a breakthrough. No specific upcoming pivotal readout was verified in this baseline. [Primary trial](https://pubmed.ncbi.nlm.nih.gov/16060712/)

#### Energy devices: comparative gains and treatment-specific safety

A February 27, 2026 randomized Chinese study compared **YOUMAGIC monopolar radiofrequency with Thermage CPT** in 230 enrolled participants, with 212 in the final analysis and six-month follow-up. The reported day-90 GAIS threshold was reached by 100% versus 98.1%; the investigators concluded noninferiority. This active-comparator design does not show superiority over sham, and percentage responders are not the percentage of loose skin removed. The registry identifier is **ChiCTR2400087993**, retrospectively registered. The abstract reported no serious adverse events; U.S. clearance of the specific YOUMAGIC device was not established in this check. [Primary study](https://pubmed.ncbi.nlm.nih.gov/41758257/)

Keep monopolar RF, RF microneedling, focused ultrasound and lasers distinct. FDA's **October 15, 2025** RF-microneedling communication describes reported burns, scarring, unwanted fat loss, disfigurement and nerve injury; it says evaluation is ongoing. The communication does not supply a complication incidence or prove all RF technologies share the same risk. Future trials should measure visible benefit alongside unintended volume loss, pigmentation changes and recovery. ### September 14, 2026 — Focused review and an older regulatory setback

**RelabotulinumtoxinA / Relfydess — July 1, 2026 FDA setback.** Galderma's [H1 2026 report](https://www.galderma.com/news/galderma-H12026-performance) reiterates a U.S. complete response letter concerning manufacturing-site observations and analytical-method optimization. This is older missing baseline context, not a new September clinical failure. Overseas approvals do not establish U.S. approval; a dependable resubmission/decision date was not verified. This neuromodulator addresses dynamic wrinkles and should not be presented as replacement of facial volume or correction of substantial laxity.

**KB304 and device safety:** the [PEARL-2 registry](https://clinicaltrials.gov/study/NCT06724900), last posted May 13, 2025, still lists active/not recruiting with elapsed 2025 completion estimates and no posted results. It does not supersede the dated sponsor results above or confirm a new trial start. The **2027 Phase 2 forecast** remains the current sponsor window. FDA's [RF-microneedling communication](https://www.fda.gov/medical-devices/safety-communications/potential-risks-certain-uses-radiofrequency-rf-microneedling-fda-safety-communication) still describes its evaluation as ongoing; no new completion date was found.

### Dated findings

**September 12, 2026 — Initial baseline.** Recorded historical human evidence, the updated KB304 development window, a 2026 device comparison and the RF-microneedling safety signal. These are baseline findings, not all newly announced today. Direct registry pages/API were incompletely retrievable; named-study evidence comes from the linked primary reports. Informal sources remain eligible under the [notebook rumor policy](../README.md#rumors-and-informal-signals); no specific new rumor was established in this selective check.

### September 14, 2026 — Focused research refresh

Checked KB304 registry/sponsor guidance, the FDA RF-microneedling communication and Galderma's portfolio report. Added the July 1 relabotulinumtoxinA complete response letter as older negative regulatory context. No new tightening or rejuvenation outcome was verified in the checked sources. Comparative-device and informal-source coverage remains selective; this focused review did not refresh every study in the category.

### 2026-09-18 — Scheduled focused review

Searched human skin-quality, tightening, regenerative and device developments, including medical-weight-loss populations. Appraised Sep 17 AbbVie sponsor announcement. Refreshed original KB304 registry and FDA radiofrequency microneedling communication; no new FDA update was found on the existing safety notice. Original event dates, safety and limitations are retained in the September 18 update above. The complete-topic successful-check date was not advanced because the listed gaps remain. [Run record](../updates/2026-09-18_scheduled_research_update.md).
