# September 14, 2026 — Ad-hoc research update

[Home](../README.md) · [Latest highlights](../LATEST.md)

Focused review of all 28 topics, using a seven-day overlap (September 5–14) and selected older missing context. Source and event dates remain distinct. Every topic received a focused search, but this was not an exhaustive re-review of every program, registry, paper or informal source. Full-topic successful-check timestamps are preserved. The Tuesday/Friday 9 a.m. Eastern schedule is unchanged.

## Findings added

### 2026-09-14 — AP-SA02 phage therapy receives FDA Breakthrough Therapy designation

**Original sponsor release.** Armata announced on September 14 that FDA granted Breakthrough Therapy designation to intravenous AP-SA02, a fixed multi-phage treatment being developed alongside antibiotics for complicated MSSA or MRSA bloodstream infections. The designation builds on the existing phase 1b/2a diSArm study (NCT05184764); it is not a new efficacy readout or an approval. Armata still anticipates initiating a phase 3 superiority study in the second half of 2026. That start remains a forecast. Larger confirmatory results are needed to establish benefit and safety beyond the small earlier trial. ### 2026-09-14 — Enhertu improves first-line HER2-mutant lung-cancer PFS; survival benefit unproven and fatal lung toxicity reported

**Original sponsor conference report.** In DESTINY-Lung04 (NCT05048797), 454 patients with untreated advanced HER2-mutant nonsquamous NSCLC were randomized to Enhertu or platinum/pemetrexed plus pembrolizumab. Sponsor-reported median progression-free survival was 14.3 versus 8.3 months (HR 0.63, 95% CI 0.50–0.79). Overall survival was immature and not formally tested: 29.3 versus 33.1 months (HR 1.15, 95% CI 0.88–1.52), so this readout does not demonstrate longer survival. Interstitial lung disease/pneumonitis occurred in 20.8% with Enhertu, including four grade 5 events (1.8%). This first-line use remains investigational in the US; prior approval for previously treated disease should not be generalized to this setting. ### 2026-09-14 — ADAURA reports sustained eight-year survival difference after adjuvant osimertinib

**Original sponsor report; linked journal article access failed.** An exploratory eight-year ADAURA follow-up reported overall survival of 74% with adjuvant osimertinib versus 58% with placebo in the resected EGFR-mutant stage II–IIIA population (HR 0.53, 95% CI 0.38–0.75). Across stages IB–IIIA, eight-year survival was 79% versus 64% (HR 0.52, 0.39–0.71). The randomized trial enrolled 682 patients. This later analysis has a follow-up limitation: 127 patients supplied no additional survival data beyond the prior planned final analysis and remained censored. It supports durability of the observed benefit but is exploratory follow-up, not a new approval or a new randomized trial. No new safety collection was reported for this analysis. ### 2026-09-14 — DT120 reports positive phase 3 anxiety results

**sponsor_topline_randomized_phase3.** **September 14, 2026 — DT120 Panorama reports a second positive Phase 3 GAD trial.** In this sponsor topline from Panorama (NCT06809595), 245 adults were randomized to lysergide ODT 100 micrograms (96), 50 micrograms (52), or placebo (97). At 12 weeks, the 100-microgram arm's HAM-A anxiety-score change was −9.8 versus −4.7 with placebo: a −5.1-point difference (p<0.0001; Cohen's d=0.64). The sponsor reports that the primary and all key secondary endpoints were met; confidence intervals were not supplied. Treatment-emergent adverse events occurred in 94.8% versus 62.9%; common dosing-day events included perceptual illusions, nausea and headache. No drug-related serious adverse event was reported. Recognizable effects can compromise masking, and the 50-microgram arm was not powered for comparisons. The 12-week controlled result does not establish permanent benefit or superiority to existing medicines; the extension allows open-label retreatment. DT120 remains investigational. Next: a pre-NDA meeting in Q4 2026 and intended filing in H1 2027, both sponsor milestones rather than approval dates. ### 2026-09-11 — Apitegromab gains an SMA approval

**Regulatory action.** FDA approved Isembyld (apitegromab-mstn) on September 11 for SMA in adults and children aged at least two already receiving an SMN2-targeted treatment. SAPPHIRE (NCT05156320) enrolled 188 participants aged 2–21; its 52-week motor-function evidence is separate from EMBRAZE’s obesity body-composition results. FDA highlights fracture risk, possible fetal harm and reproductive effects. **This does not approve muscle preservation during GLP-1 treatment.** [FDA action](https://www.fda.gov/drugs/news-events-human-drugs/fda-approves-first-therapy-target-muscle-loss-spinal-muscular-atrophy). The sponsor reported launch underway, with shipping expected in coming days; actual delivery was not independently verified. [September 11 sponsor announcement](https://investors.scholarrock.com/news-releases/news-release-details/scholar-rock-announces-fda-approval-isembyldtm-apitegromab-mstn). This supersedes the previously pending September 30 action target. ### 2026-09-11 — Camizestrant misses primary endpoint in upfront first-line SERENA-4

**Original sponsor release.** AstraZeneca reported that SERENA-4 did not achieve a statistically significant progression-free survival improvement for camizestrant plus palbociclib versus anastrozole plus palbociclib in untreated advanced ER-positive/HER2-negative breast cancer. The randomized, double-blind phase 3 trial enrolled 1,371 patients. Numerical effect estimates and confidence intervals have not been released; the sponsor reported no new safety concern. This negative result concerns upfront first-line treatment. It should not be conflated with the different SERENA-6 strategy of switching after an emerging ESR1 mutation during endocrine therapy before clinical progression. Full results remain to be presented. [Original source](https://www.astrazeneca.com/media-centre/press-releases/2026/update-serena-4-phase-iii-trial.html) · [Topic notes](../topics/14_cancer_vaccines.md).

### 2026-09-10 — Stroke polygenic scores show small, ancestry-dependent gains in external validation

**Original peer-reviewed human validation study.** A multi-cohort study evaluated function-informed and ancestry-specific ischemic-stroke polygenic risk scores. In All of Us participants of European ancestry, the strongest score had an odds ratio of 1.16 per standard deviation (95% CI 1.12–1.19), but adding scores moved discrimination only from about 66.0% to 66.3–66.6%. In the African-ancestry group, performance gains were negligible; the corresponding score association was 0.99 (0.94–1.05). This is model validation, not proof that PRS-guided screening or treatment improves health outcomes. It reinforces the need for ancestry-specific validation and modest expectations for incremental prediction. [Original source](https://www.frontiersin.org/journals/bioinformatics/articles/10.3389/fbinf.2026.1875388/full) · [Topic notes](../topics/28_genetic_screening.md).

### 2026-09-10 — CRAVE registry confirms recruitment

**trial_registry_update.** **September 14 check — CRAVE recruitment status clarified.** The September 10 ClinicalTrials.gov update now lists the VA's semaglutide alcohol-use-disorder study CRAVE (NCT07218354) as recruiting, with an actual study start of July 28, 2026. This resolves the earlier stale recruitment snapshot, without verifying every site's enrollment status. The randomized, blinded Phase 3 study plans 622 veterans; its primary outcome is at least a two-level reduction in WHO drinking-risk category over the final 28 treatment days. It has not reported efficacy or safety results and does not establish an AUD indication. Primary completion is estimated March 31, 2029, with overall completion March 30, 2030; neither is a promised publication date. ### 2026-09-10 — A small postoperative suzetrigine comparison

**conference_abstract_retrospective_historical_control.** **September 10, 2026 — preliminary postoperative opioid-use evidence.** PAINWeek abstract P062 reports a single-center retrospective comparison of 104 knee-arthroplasty patients: 52 prescribed suzetrigine within multimodal care and 52 historical controls. No opioid refill was needed by two weeks in 57.7% versus 34.6% (p=.018); 88.5% versus 65.4% were opioid-free at six weeks (p=.005). The latter comparison was not significant in the opioid-naive subgroup (p=.054). Pain and functional recovery did not differ significantly. Historical controls, baseline opioid-use differences and prescribing/dispensing uncertainties prevent a causal opioid-sparing conclusion. The abstract supplies no detailed comparative safety analysis. This is an early real-world conference signal, not a new indication or proof of chronic neuropathic-pain benefit. Prospective controlled replication is needed; no dated next result was identified. [Original source](https://painweek2026.eventscribe.net/fsPopup.asp?PresentationID=1878801&efp=QUFKU0NVSUQyNzE4MA&mode=presInfo&rnd=0.2075611) · [Topic notes](../topics/16_pain.md).

### 2026-09-09 — Oral afimetoran meets phase 2 lupus endpoint; full effect sizes pending

**Original sponsor release and registry.** Beeline reported positive phase 2 results for afimetoran, an oral TLR7/8 inhibitor, in 248 adults with active moderate-to-severe systemic lupus erythematosus. In NCT04895696, three daily doses were compared with placebo for 48 weeks alongside background therapy and a required corticosteroid taper. All three doses improved the week-48 SRI-4 response endpoint versus placebo (p<0.001 for each). Response percentages, effect sizes and detailed adverse-event tables were not released. The sponsor reported no new safety signal and plans pivotal development. This is a sponsor topline result, not evidence of a cure or an FDA approval; full data are needed to judge clinical magnitude. [Original source](https://beelinemedicines.com/press-releases/beeline-medicines-announces-positive-topline-results-from-global-phase-2-study-of-afimetoran-in-systemic-lupus-erythematosus/) · [Topic notes](../topics/15_autoimmune.md).

### 2026-09-09 — Oveporexton phase 3 trials reach publication

**sponsor_report_of_peer_reviewed_publication.** **September 9, 2026 — pivotal oveporexton evidence reaches journal publication.** Takeda announced NEJM publication of FirstLight and RadiantLight, the 12-week placebo-controlled narcolepsy type 1 trials already underlying the approval record (168 and 105 participants). The publication adds a peer-reviewed reporting milestone for wakefulness, sleepiness and cataplexy, with additional quality-of-life analyses. Frequent transient insomnia and urinary symptoms remain important; short controlled follow-up does not establish long-term comparative benefit. The sponsor announcement and indexed journal abstract were checked; full text and supplements could not be retrieved for independent detailed appraisal. This is a publication update, not a new approval. The official U.S. product page still says availability is expected by November 2026, pending DEA scheduling, when checked September 14. [Original source](https://www.takeda.com/newsroom/newsreleases/2026/nejm-oveporexton-narcolepsy-data/) · [Topic notes](../topics/12_sleep.md).

### 2026-09-08 — HARBOR misses its primary muscle-function endpoint

**Sponsor topline — negative result.** Novartis reported on September 8 that del-desiran’s phase 3 HARBOR trial in myotonic dystrophy type 1 missed its primary video hand-opening-time endpoint. Favorable secondary/exploratory signals do not overturn that negative primary result. Detailed estimates and safety tables remain unavailable; the sponsor reported safety consistent with earlier data and will review the development path with regulators. [Sponsor report](https://www.novartis.com/us-en/news/media-releases/novartis-provides-update-delpacibart-etedesiran-del-desiran-phase-iii-harbor-study-treatment-myotonic-dystrophy-type-1-dm1). The [NCT06411288 registry](https://clinicaltrials.gov/study/NCT06411288) records 159 actual participants and July 23 primary completion; this is disease-specific evidence, not a general muscle-building study. No approval or dependable next-results date was established. ### 2026-09-08 — Tozorakimab reduces COPD exacerbations in two phase 3 trials; US review underway

**Peer-reviewed abstract retrieved via Europe PMC; original sponsor release.** Full OBERON and TITANIA results evaluated tozorakimab 300 mg every four weeks for 52 weeks in COPD with prior exacerbations despite inhaled maintenance therapy. In the primary former-smoker population, annualized moderate/severe exacerbations were 1.34 versus 1.90 in OBERON (rate ratio 0.71, 95% CI 0.57–0.88) and 1.37 versus 2.07 in TITANIA (0.66, 0.55–0.80). Overall adverse-event frequencies were 70.4% versus 77.2% and 80.1% versus 79.8%, respectively. These are exacerbation outcomes, not mortality evidence. AstraZeneca also reported FDA Priority Review of the COPD application, with an anticipated decision during Q1 2027. Tozorakimab remains investigational. [Sponsor regulatory announcement](https://www.astrazeneca.com/media-centre/press-releases/2026/tozorakimab-demonstrated-statistically-significant-highly-clinically-meaningful-reduction-copd-exacerbations-oberon-titania-phase-iii-trials.html). ### 2026-09-08 — DeLLphi-305 maintenance combination meets survival endpoint in extensive-stage SCLC

**Original sponsor release.** AstraZeneca reported that adding tarlatamab to durvalumab maintenance improved overall survival, progression-free survival and response rate versus durvalumab alone in phase 3 DeLLphi-305. The open-label trial randomized 563 patients with extensive-stage small-cell lung cancer without progression after durvalumab plus platinum/etoposide induction. Hazard ratios, absolute survival estimates and detailed safety results were not released; the sponsor reported no new safety signal. Full results are required to assess the size and trade-offs of benefit. This report does not establish US approval of the maintenance combination. ### 2026-09-08 — An oral DME treatment enters phase 2

**sponsor_trial_initiation_with_registry.** **September 8, 2026 — oral BGE-102 begins a vision-outcome trial in diabetic macular edema.** BioAge announced first dosing in QUELL-DME (NCT07812090), a randomized Phase 2 study planning 180 adults. The three groups compare anti-VEGF injection plus oral placebo, anti-VEGF plus BGE-102, and BGE-102 with sham injection. The primary endpoint is best-corrected visual acuity at 12 weeks; retinal and inflammatory biomarkers are exploratory. There are no DME efficacy or comparative safety results yet, and this does not establish benefit for geographic atrophy. BGE-102 is investigational. The current registry lists recruiting, an actual August 21 study start and estimated November 2027 primary completion; BioAge forecasts topline results in H2 2027. Registry status does not establish that every listed site is enrolling. [Original source](https://rss.globenewswire.com/news-release/2026/09/08/3357592/0/en/bioage-labs-announces-first-participant-dosed-in-quell-dme-a-phase-2-trial-of-bge-102-a-novel-oral-nlrp3-inhibitor-in-diabetic-macular-edema.html) · [Topic notes](../topics/18_hearing_vision.md).

### 2026-09-07 — Pediatric semaglutide trial reports positive topline

**Sponsor topline — detailed results pending.** Novo’s September 7 release reports that STEP Young met its 68-week BMI primary endpoint in 165 children aged 6 to under 12 with obesity. Under the trial-product estimand, 40.4% versus 0% crossed below the age- and sex-specific obesity threshold. That is a secondary classification endpoint, not percentage weight loss. Numerical primary-endpoint effects, intervals and detailed adverse-event tables were not released; the sponsor reported no new safety or growth/puberty concerns. **No new approval for this age group is established by these results.** Full data are scheduled for ObesityWeek, November 14–17. [Sponsor topline](https://www.novonordisk.com/news-and-media/news-and-ir-materials/news-details.html?id=916600). [NCT05726227](https://clinicaltrials.gov/study/NCT05726227) includes children and adolescents and still lists estimated enrollment of 210; that wider registry count must not replace the reported younger-cohort denominator. ### 2026-09-06 — Cell-enhanced grafting improves scar scores in reconstructive wounds

**Peer-reviewed randomized human study.** September 6, 2026 — Randomized reconstructive-skin study. In 232 patients with full-thickness defects, adding an autologous epidermal-stem-cell-enriched suspension to composite skin grafting improved six-month clinician-rated scar quality (median Vancouver Scar Scale 3 versus 6). Early wound closure also favored the intervention after statistical adjustment; wound recurrence did not significantly differ. The single-center study analyzed six-month completers and lacked patient-reported outcomes. Authors reported no treatment-related adverse events through six months. This concerns wounds requiring reconstruction, not disappearance of ordinary established cosmetic scars; U.S. authorization for this protocol was not established. [Primary paper](https://doi.org/10.1002/mco2.70973). [Original source](https://doi.org/10.1002/mco2.70973) · [Topic notes](../topics/21_scars.md).

### 2026-09-02 — Older context added: NATiV3 reaches final patient visit

**Sponsor development milestone — baseline addition.** Inventiva’s September 2 announcement says the last NATiV3 participant completed the 72-week visit. The main cohort included 1,009 adults with noncirrhotic MASH and F2–F3 fibrosis, plus 410 exploratory participants. **No efficacy or safety results were reported.** Topline is sponsor-targeted for Q4 2026; a possible H1 2027 filing depends on results. [Sponsor announcement](https://inventivapharma.com/wp-content/uploads/Inventiva-PR-Inventiva-LVLP-EN-09-02-2026.pdf). The July 2 [NCT04849728 registry](https://clinicaltrials.gov/study/NCT04849728) retains a September 2027 completion estimate encompassing an additional 48-week extension safety follow-up; this is distinct from the 72-week topline milestone. Its estimated enrollment of 1,000 differs from the later sponsor counts. Neither registry completion nor the final visit guarantees a publication date. Added September 14 as missing earlier context, not same-day news. ### 2026-09-02 — Older context added: a small combination geroscience pilot

**Sponsor preliminary human report — baseline addition.** AgelessRx’s September 2 report describes a randomized 90-day combination pilot: 26 enrolled and 18 analyzed across three small groups. Reported biomarker, lean-mass and quality-of-life signals are exploratory. Multiple drugs and behavioral components changed together; attrition, small samples, missing between-group uncertainty and sponsor involvement limit inference. **This does not establish lifespan extension or isolate any drug’s benefit.** [Sponsor report](https://agelessrx.com/agelessrx-xprize-pilot-longevity-protocol-results/); [NCT07475546](https://clinicaltrials.gov/study/NCT07475546), which still has no posted results. Separately, [RESTOR / NCT06658093](https://clinicaltrials.gov/study/NCT06658093) lists recruiting and estimated July 2028 primary completion; that is not a publication date. AFAR still describes [TAME fundraising to launch](https://www.afar.org/tame-trial). These are focused baseline/status additions, not proof of rejuvenation. ### 2026-08-24 / confirmed by 2026-08-31 — Earlier safety update: rap-cel halt and separate zola-cel enrollment pause

**Original reporting with emailed company confirmations; standalone sponsor notice not retrieved.** In company statements reported by BioPharma Dive on August 31 and updated September 1, Novartis confirmed that it halted rap-cel autoimmune/neurology studies on August 24 after three cases of immune effector cell-associated hemophagocytic syndrome led to fatal complications. BMS separately confirmed a voluntary pause in zola-cel autoimmune enrollment after transient, reversible inflammatory events. These are distinct programs and events; the reports do not establish a class-wide failure. No restart date was confirmed in the sources checked. This corrects missing late-August safety context rather than representing new September 14 news. [Original source](https://www.biopharmadive.com/news/novartis-bristol-myers-autoimmune-cell-therapy-trial-halt/829218/) · [Topic notes](../topics/15_autoimmune.md).

## Other baseline and registry work

The skin, hair and dental notes add explicitly older ABS201 safety/PK context, the July relabotulinumtoxinA manufacturing-related FDA complete response letter, May Belotero indication approval and May Differin Epiduo OTC switch. These are not September 14 news. Registry refreshes address acne-vaccine, verteporfin, vitiligo and orthodontic studies; a completed or elapsed trial date does not establish a result.
