Weight loss

REDEFINE 1 monotherapy comparator

11.5% reported weight loss · 3% loss with placebo · 68 weeks

2.4 mg once weekly

Adults with obesity, or overweight plus a weight-related condition, without diabetes

Effect regardless of treatment discontinuation or rescue weight-management treatment Investigational; RENEW phase 3 monotherapy programme underway

Safety context

Arm-specific safety not extracted; CagriSema combination rates must not be assigned to monotherapy.

Limitations: Different populations, durations and titration schedules prevent causal cross-trial ranking.

Glucose control

REIMAGINE 2

1.02 percentage-point HbA1c decrease · 0.62 pp decrease with Placebo · 68 weeks

2.4 mg weekly

Adults with type 2 diabetes

Treatment-regimen estimate, regardless of treatment discontinuation or additional glucose-lowering therapy.

Safety context

GI events 39.5%; adverse-event discontinuation 4.6%.

Limitations: Trial-wide baseline. Results do not establish superiority across unrelated trials.

Hormone activity

An amylin analogue that activates amylin receptors and also activates calcitonin receptors.

Investigational monotherapy (formerly AM833); no approved commercial brand verified. CagriSema is a separate combination development program.

Novo Nordisk · Developer of cagrilintide monotherapy and its CagriSema combination program.

Uses, doses and availability depend on the formulation, indication and country. Study doses below describe research; they are not prescribing instructions.

The molecule

Experimental fragment

Cagrilintide · receptor-bound peptide model

Ribbon representation of the experimentally modeled cagrilintide peptide chain, extracted by RCSB from its amylin-2 receptor complex.

Shown: Receptor-bound peptide model; a partial representation of the complete modified drug.

Cryo-EM, 2.80 Å. This ribbon shows the modeled peptide backbone and disulfide; it does not depict every atom or the full C20 lipid/linker. The receptor and G proteins are omitted. It represents one bound experimental conformation, not the sole shape in solution.

  1. Phase 2 monotherapy study published

A 26-week dose-finding study compared cagrilintide with placebo and liraglutide in adults without diabetes.

    Source
  2. Phase 3 monotherapy comparator reported

The REDEFINE 1 publication included a randomized cagrilintide-alone arm alongside the combination, semaglutide and placebo arms.

  3. RENEW phase 3 program reported underway

Novo Nordisk's 2025 annual report, released in February 2026, reported initiation of RENEW 1 and RENEW 2 for obesity with and without type 2 diabetes. This dates the status disclosure, not the first participant.
