Weight & glucose

Eloralintide

Preferentially activates amylin receptors. Laboratory studies also found weaker calcitonin receptor activity; the targets do not have equal potency, and their separate contributions to weight loss are not established.

Weight loss

48-week phase 2

20.1% reported weight loss · 0.4% loss with placebo · 48 weeks

9 mg once weekly

Adults with obesity, or overweight plus a weight-related condition, without diabetes

Efficacy estimand; endpoint not adjusted for multiplicity Investigational; ENLIGHTEN phase 3 programme underway

Safety context

GI symptoms and fatigue were more frequent at higher doses; slower escalation improved tolerability.

Limitations: Efficacy estimand only; 9 mg arm is small. Paper abstract rounds loss to 20%; sponsor reports 20.1%.

Glucose control

Eloralintide monotherapy arm in EloraTZP phase 2b

1.4 percentage-point HbA1c decrease · 0.3 pp decrease with Placebo · 48 weeks

6 mg once weekly after escalation

Adults with obesity or overweight and type 2 diabetes

Efficacy estimand assumes continued intervention through 48 weeks, allowing dose interruptions/modifications. Values rounded to one decimal in the sponsor release.

Safety context

GI adverse events predominated. Adverse-event discontinuation ranged from 0% to 10.8% across eloralintide monotherapy arms; the retrieved release does not assign that range to the 6 mg arm.

Limitations: This is a monotherapy arm of a 367-person multi-arm study, not 367 people receiving 6 mg. The 6 mg arm had the largest reported monotherapy HbA1c decrease; the 9 mg arm decreased by 1.3 pp. No durable diabetes-complication outcome or U.S. approval established.

  4. Announcement

Diabetes monotherapy and EloraTZP combination readout

The NCT06603571 monotherapy 6 mg arm decreased HbA1c 1.4 pp versus 0.3 pp with placebo and weight 12.3% versus 3.0% at 48 weeks (rounded efficacy estimates). Separate eloralintide plus tirzepatide arms supported a planned co-formulated phase 3 program; these are distinct from the earlier non-diabetic monotherapy study.
