Eloralintide activates amylin receptors; tirzepatide activates GLP-1 and GIP receptors. The phase 2b study used separate injections. A co-formulated product is planned for phase 3; minimal calcitonin-receptor activity is not presented as an equal therapeutic target.
At eloralintide 9 mg + tirzepatide 15 mg: nausea 49%, diarrhea 30%, vomiting 24%; 27% discontinued because of adverse events. Comparator discontinuation was 2.9% with tirzepatide and 16.7% with placebo (slides round to 3% and 17%). Across combination arms, discontinuation ranged from 10.8% to 27.0%.
Eloralintide 9 mg + tirzepatide 15 mg once weekly, separate injections
Adults with obesity or overweight and type 2 diabetes
Model-based efficacy estimand: assumes all randomized participants remained on intervention through week 48, allowing dose interruptions/modifications. The modeled endpoint is separate from observed-visit means. Investigational; co-formulated AMPLIFY phase 3 planned for Q4 2026, not confirmed started.
Safety context
At eloralintide 9 mg + tirzepatide 15 mg: nausea 49%, diarrhea 30%, vomiting 24%; 27% discontinued because of adverse events. Comparator discontinuation was 2.9% with tirzepatide and 16.7% with placebo (slides round to 3% and 17%). Across combination arms, discontinuation ranged from 10.8% to 27.0%.
Limitations: Small phase 2b arms; sponsor-reported efficacy estimand assumes continued intervention despite the substantial discontinuation rate. Primary comparison was weight change versus placebo; HbA1c and tirzepatide comparisons were secondary. Placebo HbA1c decrease was 0.29 pp. Weight loss was 23.3% versus 14.8% with tirzepatide and 3.0% with placebo. No co-formulated phase 3 result or approval.

Selected combination-specific milestones. Earlier development of each component is recorded under eloralintide and tirzepatide; this is not a complete development history.
Announcement
Phase 2b results and phase 3 plan announced
In 367 adults with obesity/overweight and T2D, the 9 mg + 15 mg combination achieved 23.3% weight loss and 2.91 pp HbA1c reduction at 48 weeks (efficacy estimand). Tirzepatide achieved 14.8% and 2.41 pp; placebo 3.0% and 0.29 pp. AE discontinuation was 27% at this combination dose. Co-formulated AMPLIFY phase 3 is forecast for Q4 2026, not a confirmed start.
