Weight loss

ATTAIN-1

11.2% reported weight loss · 2.1% loss with placebo · 72 weeks

36 mg once-daily trial capsule (bioequivalent approved tablet: 17.2 mg)

Adults with obesity, or overweight plus a weight-related condition, without diabetes

Published treatment-regimen estimand in the intention-to-treat population FDA-approved April 1, 2026 for adult weight management (Foundayo)

Safety context

GI effects predominated; adverse-event discontinuation at top dose 10.3%.

Limitations: Published trial estimate is 11.2%; subsequent label/approval communication reports 11.1%. Capsule and tablet milligram strengths differ.

Glucose control

ACHIEVE 3

1.91 percentage-point HbA1c decrease · 1.47 pp decrease with Oral semaglutide 14 mg · 52 weeks

  5. Announcement

Orforglipron comparison extends to oral semaglutide 25 mg

A covariate-adjusted indirect analysis compares orforglipron 17.2 mg against oral semaglutide 25 mg in adults with type 2 diabetes. This is not the ACHIEVE-3 randomized comparison against oral semaglutide 14 mg. Cross-study assumptions and treatment-adherent estimands limit causal interpretation.
