Lipid lowering
How to read this chart
What changed in the lipid measurement?
- Choose the marker first, then the comparison: change from baseline and difference versus placebo answer different questions.
- Check dose, timing and study population. A lower biomarker does not by itself establish fewer cardiovascular events.
- Select a result for its definition and uncertainty. Derived within-arm values do not borrow confidence intervals from placebo-adjusted estimates.
Terms in this view
- Confidence interval
- A range expressing statistical uncertainty around an estimate. It is not the range of individual patients’ outcomes. A 95% interval procedure covers the true value in about 95% of repeated studies under its assumptions. Reference
- Absolute versus relative
- A fall from 10% to 8% is 2 percentage points lower, or a 20% relative reduction. The starting value and follow-up period matter. Reference
Figures include sources, study context and the current selection.
Cholesterol carried by LDL particles.
Positive values indicate lowering; negative values indicate an increase. Populations, baseline levels, and analysis methods differ; lipid changes do not establish event reduction.

Study selection & comparison notes
Representative sourced regimens from established and investigational lipid therapies. This is a growing evidence library, not an exhaustive ranking or a network meta-analysis. Dose, population, background treatment and follow-up differ across trials.
Change from baseline and difference versus placebo are separate views. When an arm change is supplied alongside an adjusted contrast, the baseline view uses that reported arm change; the adjusted confidence interval is not reused. Missing values are not treated as zero.
Positive values mean lipid lowering; negative values mean an increase. All extracted intervals are 95% confidence intervals. Mean and median estimates remain labeled; their methods are not interchangeable. Increasing HDL-C is not assumed to reduce cardiovascular risk.
LDL-C, non-HDL-C and ApoB describe different aspects of atherogenic lipoproteins. Biomarker lowering does not by itself demonstrate fewer cardiovascular events or deaths; use the separate outcomes page for those results.
Time-averaged responses remain in the therapy comparison but are excluded from the treatment-duration map. VERVE-102 averages participant responses from day 28 onward; lepodisiran averages days 60–180; selected evolocumab markers average weeks 10 and 12.
Time courses connect only extracted visits and mark approximate figure readings with hollow points. Studies with only two follow-up estimates are displayed as disconnected points with confidence intervals. No missing visits or trajectories are modeled.
Lp(a) assays and units differ. No fixed conversion is applied between nmol/L and mg/dL. A placebo-adjusted reduction may exceed 100% when placebo levels rise; this does not imply a negative blood concentration. Muvalaplin results here use the intact-Lp(a) assay.
Label safety summaries are distinguished from trial-specific findings. Rare-disease cohorts, small dose groups, and source-specific statistical limitations are described in study details.
Coverage & missing results
- VERVE-101
Earlier Heart-1 proof-of-concept program; LDL fell 46% in five evaluable participants at 0.45 mg/kg, but enrollment paused after liver-enzyme and platelet abnormalities. VERVE-102 is the current plotted successor.
Source - Inclisiran time course
An endpoint is included. Its public figure and imputed endpoint table use analyses that need reconciliation before a curve can be plotted.
Source - ARO-DIMER-PA
September 15 sponsor interim results report dual PCSK9/ANGPTL3 RNAi lipid lowering. Complete dose-specific denominators and comparable time points were not verified; reported maxima are not plotted as comparable mean endpoints. No cardiovascular-event benefit established.
