September 12, 2026 — initial evidence brief and cover page
Requested work: establish a current-state brief for the original eight categories, add expected-update timing, identify the cover page, and add psilocybin/related therapies as a ninth category.
Deliverables: cover page, dated initial brief, watchlist, and the topic notes below. This is the first focused baseline, not an exhaustive systematic review. Results before today are baseline context, not presented as newly occurring news.
| Topic | Evidence covered | Main point / next trigger |
|---|---|---|
| Whole-body MRI | Professional society positions, HERCULES investigator abstract and study site, UK Biobank project, new Cochrane protocol | No demonstrated population mortality benefit identified; no reliable definitive readout date. |
| Galleri | NHS-Galleri and PATHFINDER 2 results, REACH and Vanguard context, FDA panel announcement | Missed primary endpoint; September 23 FDA panel is next dated event. |
| GLP-1 | FDA approvals, six major pipeline programs, pivotal and comparative results, sponsor timelines | Oral options already approved; EASD and late-2026/2027 development milestones. |
| Muscle | BELIEVE and EMBRAZE papers, COURAGE results/safety, SRK-439 and enobosarm programs | Lean-mass preservation is better established than functional benefit. Late-2026 and Q1-2027 sponsor windows. |
| Longevity | AFAR TAME status, metformin trials/preprint, rapamycin and senolytic randomized trials | TAME launch unconfirmed; mixed clinical evidence and no credible lifespan-results timetable. |
| CBL-514 | Full Phase 2b paper, SUPREME-01 registry snapshot, updated sponsor trial announcements | Local fat signal with attrition and tolerability limitations; September 30 presentation and provisional 2027 pivotal window. |
| Cardiovascular | VERVE/Lilly, enlicitide FDA approval, pelacarsen negative topline, Lp(a) outcomes timelines, olezarsen expansion | Full HORIZON results and VERVE phase 2; later outcomes trials extend into 2028–2029. |
| Alzheimer's | Diagnostic FDA decisions, Leqembi/Kisunla, prevention studies, trontinemab and diranersen | Diagnostic use versus screening kept separate; larger prevention/pivotal outcomes in 2027–2028 estimates. |
| Psychedelics | Psilocybin and related clinical programs, current FDA guidance/hearing, sponsor readouts and filing plans | September 14 FDA hearing; compound-specific efficacy and regulatory milestones, without treating filings as approvals. |
Coverage and interpretation limits
- Primary papers, regulators, trial teams, registries and dated sponsor releases support the notes. Company-only results and conference reports are labeled.
- MRI/HERCULES and SUPREME registry detail/API rendering was unreliable. Alternate primary study sites, investigator abstracts, the indexed primary registry snapshot and newer dated sponsor statements provided the baseline; unresolved registry freshness is explicitly recorded.
- Full NHS-Galleri and PATHFINDER 2 peer-reviewed outcome publications/subgroup tables were not appraised. Their baseline summaries retain trial-team/sponsor-conference attribution.
- Primary completion, overall study completion, readout, regulatory submission and approval are different events. Dates in the cover-page table preserve these differences. No deadline is invented where no reliable source exists.
- Independent review checked the key BELIEVE/EMBRAZE lean-mass and functional findings and COURAGE safety wording against the primary sources; no material discrepancy was found.
Recurring checks
The active Tuesday/Friday automation already reads the evolving watchlist as authoritative, including additions. The ninth category therefore joins the existing schedule without creating another task. Future runs should compare against these saved baselines, retain dated history, and refresh the cover page when current status or expected timing changes.
Verification completed at 2026-09-12 20:04 UTC: all nine topic briefs read back, all nine initial-brief sections present, all local Markdown links resolved, and state.json persisted with nine completed focused baselines and explicit coverage notes. The existing automation was read back as active and configured to follow the evolving watchlist.
