Weight-loss trials

How to read this chart

How much weight was lost, and over how long?

  1. Read weeks along the bottom and average body weight lost on the vertical axis. The points are study readouts, not predictions for an individual.
  2. Check population, dose and analysis before comparing. Different trials are not head-to-head comparisons. Curves can use a different analysis from the endpoint map.
  3. Select a point for its comparator, safety and source. No uncertainty bar plotted does not mean there is no uncertainty.
Terms in this view
Confidence interval
A range expressing statistical uncertainty around an estimate. It is not the range of individual patients’ outcomes. A 95% interval procedure covers the true value in about 95% of repeated studies under its assumptions. Reference
Absolute versus relative
A fall from 10% to 8% is 2 percentage points lower, or a 20% relative reduction. The starting value and follow-up period matter. Reference
Treatment-policy analysis
Estimates the effect of assigning treatment while including outcomes after specified events such as stopping treatment or using rescue medication. Missing outcomes may still require assumptions. Reference
Efficacy / hypothetical analysis
Often estimates what would happen if participants continued treatment without specified interruptions or rescue therapy. Check the trial definition: this differs from treatment policy and is not automatically a completers-only analysis. Reference

Figures include sources, study context and the current selection.

Incretin mimetics, such as GLP-1s, and related weight-loss therapies.

Hormone targets

Includes therapies acting on any selected hormone’s receptors. Meet the hormones.

What is included—and how to compare it

This snapshot covers publicly reported adult weight-management trials of GLP-1 medicines, dual and triple agonists, and related amylin therapies. Approved medicines provide context for the investigational pipeline. Diabetes-selected, pediatric, MASH-selected, and withdrawal-maintenance studies are not mixed into the main comparison. Mazdutide 9 mg (marked *) is a reported non-diabetic subgroup of a mixed-population trial.

Each point is one selected study and dose, not a pooled estimate or a drug’s maximum possible effect. A larger general-obesity randomized trial is preferred when usable results exist; within it, the highest planned maintenance dose is normally selected. Separate oral formulations, weekly versus monthly berobenatide regimens, and clinically distinct semaglutide and mazdutide dose programs remain separate. Petrelintide uses the sponsor-designated maximally effective regimen because the release does not specify its milligram dose. CT-996 and ABBV-295 are also retained as explicitly dose-undisclosed early cohorts.

The map prefers treatment-policy or treatment-regimen estimates, which include the consequences of stopping treatment. If unavailable, it uses a clearly labeled efficacy or other analysis. The analysis filter separates these. Older carried-forward analyses, small cohort means, per-protocol results, and incompletely described sponsor estimates are grouped as Other. They are not equivalent to one another.

Results are absolute average percentage losses from starting weight. Placebo values are reported separately; missing placebo values remain missing. A negative placebo loss means weight gain. Trial length, population, dose escalation, lifestyle support, treatment discontinuation and missing-data methods all matter. This plot cannot establish which drug is superior or safer. Uncertainty intervals are not harmonized or displayed; their absence does not imply exactness.

Curves use actual reported visits, from source spreadsheets, printed numerical labels, or explicitly labeled estimates read from published figures. Straight segments connect those visits without fitting or extrapolating. Observed means among remaining participants can differ from a trial’s modeled endpoint. Curve doses and trials can also differ from the endpoint map; each retains its own labels and sources. Curve coverage is curated: absence here does not mean that no published time-course figure exists for that therapy.

Safety summaries are brief study-specific context, not a complete side-effect comparison. Source papers, regulatory labels and trial registries remain the references for detailed safety. Sponsor reports are identified separately from peer-reviewed publications. Development status is a dated research snapshot and does not establish local availability.

Incretins include GLP-1 and GIP. Related amylin therapies are included in this comparison, but amylin is a different pancreatic hormone rather than an incretin. Shape encodes the phase of the plotted study, not a compound’s latest development stage. Hormone filters include any selected receptor target, whether activated or blocked. A documented secondary target does not establish its contribution to the observed weight loss.

September 22 selective addition: REDEFINE 9 lower-dose sponsor endpoints remain separate from the larger REDEFINE 1 policy estimate. No new time-course points were extracted. Other records retain earlier review dates.

October 2 selective update: EloraTZP and eloralintide diabetes results plus TRIUMPH-2 are sourced to September 29–30 reports. Diabetes-selected weight results remain outside the non-diabetic map. Existing endpoints and curves keep their historical sources; no new curve points were extracted.

Other programs & coverage gaps

This is a sourced snapshot, not a complete census of every global trial. Programs below are not assigned an invented or placebo-adjusted point.

  • HRS-1596 · weekly oral GLP-1/GIP

Phase 1 planned in 122 participants; the September 25 registry still says not yet recruiting. No human efficacy result to plot. Primary completion estimated December 2026; final completion May 2027. Checked September 29.

  • Bofanglutide · GRADUAL-1

  • EloraTZP · obesity with type 2 diabetes

Phase 2b, 48 weeks: eloralintide 9 mg + tirzepatide 15 mg produced 23.3% weight loss versus 14.8% with tirzepatide and 3.0% with placebo, using an efficacy estimand. The diabetes-selected cohort stays outside the non-diabetic map; its HbA1c result is in the glucose explorer. AMPLIFY co-formulation phase 3 is forecast for Q4 2026, not confirmed started.

    Source
  • Retatrutide · TRIUMPH-2 diabetes cohort

The September 29 Lancet report gives 18.8% mean weight loss at 80 weeks with 12 mg versus 5.1% with placebo (primary treatment-regimen analysis; n=1,152). This population had type 2 diabetes, so it is retained separately from the non-diabetic TRIUMPH-1 point. The HbA1c explorer includes its separate efficacy-estimand result.

    Source
  • Petrelintide · ZUPREME-2 diabetes cohort

October 7 ZUPREME-2 topline in type 2 diabetes (28 weeks) reports efficacy-estimand weight loss 7.4–9.2% versus 2.0% and maximum absolute HbA1c reduction 0.65 percentage points versus a 0.23-point placebo increase. Dose-to-result mapping, arm sizes and intervals are missing, so no dose-specific point or curve is plotted. Registry 221 actual participants differs from sponsor 220. The earlier non-diabetic ZUPREME-1 evidence remains unchanged.
